摘要
目的通过网络药理学方法研究中药"大黄-牡丹皮"药对治疗急性阑尾炎的作用机制。方法通过TCM-SP数据库筛选出大黄、牡丹皮的活性成分及相关靶点,在Gene Gards数据库检索急性阑尾炎的相关靶点,对两类靶点取交集,应用Cytoscape3.7.2软件构建大黄-牡丹皮治疗急性阑尾炎的成分-靶点-疾病网络,对网络中基因构建蛋白互相互作用网络并进行GO和KEGG通路富集分析。结果根据筛选条件得到化学成分27个,对应的作用靶点274个,疾病相关靶点657个。GO富集分析得到生物过程(BP)条目336个、分子功能(MF)55个、细胞组分(CC)39个,KEGG通路富集分析得到信号通路99条。结论大黄-牡丹皮治疗急性阑尾炎的主要成分有槲皮素(quercetin)、山柰酚(kaempferol)、芦荟大黄素(aloe-emodin)、β-谷甾醇(beta-sitosterol)等,通过TNF信号通路、HIF-1信号通路、Fox O信号通路、p53信号通路等,作用于IL-6、VEGFA、AKT-1、CASP-3、EGF、MAPK-8、MMP-9、PTGS-2等靶点,发挥抑制炎症、抗细胞凋亡等作用,达到治疗急性阑尾炎的目的。
Objective To study the mechanism of Dahuang(Rhei Radix Et Rhizoma)-Mudanpi(Moutan Cortex) treating acute appendicitis based on network pharmacology.Methods The TCMSP database was used to screen the effective components and corresponding targets of Dahuang(Rhei Radix Et Rhizoma)-Mudanpi(Moutan Cortex).Gene Gards database was used to predict the disease targets,and the two types of targets were mapped.The component-target-disease network of Dahuang(Rhei Radix Et Rhizoma)-Mudanpi(Moutan Cortex) for treating acute appendicitis was constructed by Cytoscape 3.7.2 software.Protein-protein interaction(PPI) network was built by string platform.GO function annotation and KEGG pathway were analyzed in David database.Results According to the screening conditions,27 components and 274 disease targets were obtained.GO enrichment analysis resulted in 336 BP entries,55 MF entries and 39 CC entries in GO entries.A total of99 KEGG signaling pathways were obtained.Conclusion The main components of Dahuang(Rhei Radix Et Rhizoma-Mudanpi(Moutan Cortex) in the treatment of acute appendicitis are quercetin,kaempferol,aloe-emodin and beta-sitosterol,through TNF signaling Pathway,HIF-1 signaling pathway,Fox O signaling pathway and p53 signaling pathway,regulating IL-6,VEGFA,AKT-1,CASP-3,EGF,MAPK-8,MMP-9,PTGS-2 and other targets.It can achieve the role of inhibiting inflammation and anti-apoptosis so as to achieve the purpose of treating acute appendicitis.
