摘要
目的检测接受高效抗逆转录病毒治疗(HAART)的人类免疫缺陷病毒(HIV)感染者/艾滋病(AIDS)患者血浆中白介素(IL)-35及其受体IL12rβ2、糖蛋白130(gp130)水平的动态变化及意义。方法选择我院2017年1月至2018年3月收治的41例HIV感染者/AIDS患者为研究对象,选择45例同期健康体检者为对照组。分别采集HIV/AIDS患者接受HAART药物0、1、6、12个月时的外周静脉血,检测血浆中的IL-35及其受体IL12rβ2、gp130水平,同时检测血浆HIV-1 RNA载量,分析IL-35与IL12rβ2、gp130、HIV-1 RNA以及IL12rβ2、gp130与HIV-1 RNA病毒载量的相关性。结果与对照组相比,接受HAART(0、1、6个月)时患者血浆中IL-35、IL12rβ2、gp130水平降低(均P<0.05)。与接受HAART 0个月相比,接受HAART 1个月时患者血浆中IL-35、gp130水平均升高,HIV-1 RNA载量降低(均P<0.05);接受HAART 6、12个月时患者血浆中IL-35、IL12rβ2、gp130水平均升高,HIV-1 RNA载量降低(均P<0.05)。随着接受HAART时间的延长(1~12个月),患者血浆IL-35、IL12rβ2、gp130水平逐渐升高,HIV-1 RNA载量逐渐降低(均P<0.05),呈时间依赖性。接受HAART 0、1个月时患者血浆IL-35与IL12rβ2呈正相关,与HIV-1 RNA载量呈负相关(均P<0.05)。接受HAART 6、12个月时患者血浆IL-35与IL12rβ2、gp130呈正相关,与HIV-1 RNA载量呈负相关(均P<0.05),同时gp130与HIV-1 RNA载量呈负相关(均P<0.05)。接受HAART 12个月时患者血浆IL12rβ2与HIV-1 RNA载量呈负相关(r=-0.457,P<0.001)。结论接受HAART的HIV/AIDS患者血浆IL-35及其受体IL12rβ2、gp130水平随着接受HAART时间的延长而逐渐升高,且与HIV-1 RNA载量关系密切。血浆IL-35及其受体IL12rβ2、gp130在抗HIV感染中可能发挥一定作用。
Objective To detect the dynamic changes and significance of the levels of interleukin-35(IL-35) and its receptors IL12 rβ2 and glycoprotein 130(gp130) in the plasma of HIV/AIDS patients who received highly active antiretroviral therapy(HAART). Methods A total of 41 HIV-infected/AIDS patients who were treated in our hospital from January 2017 to March 2018 were enrolled, and 45 cases of people who had a physical examination during the same period as the control group. The peripheral venous blood specimens were collected from HIV/AIDS patients at 0, 1, 6 and 12 months after receiving HAART, the levels of IL-35, IL12 rβ2, and gp130 in plasma were detected, and plasma HIV-1 RNA load was detected, the correlation between IL-35 and IL12 rβ2, gp130, HIV-1 RNA load, and the correlation between IL12 rβ2, gp130 and HIV-1 RNA load were analyzed. Results Compared with the control group, the plasma levels of IL-35, IL12 Rβ2 and gp130 in the patients decreased at(0, 1, and 6) months after receiving HAART(all P<0.05). Compared with for 0 month after receiving HAART, the plasma levels of IL-35 and gp130 increased at 1 month after receiving HAART, and the HIV-1 RNA load decreased(all P<0.05);the levels of IL-35, IL12 Rβ2 and gp130 increased at 6 and 12 months after receiving HAART, and the HIV-1 RNA load decreased(all P<0.05). With the prolongation of HAART time(1 to 12 months), the levels of IL-35, IL12 Rβ2 and gp130 in HIV/AIDS patients increased gradually and the HIV-1 RNA load decreased gradually(all P<0.05), in a time-dependent way. At 0 and 1 month after receiving HAART, plasma IL-35 was positively correlated with IL12 rβ2, and negatively correlated with HIV-1 RNA load(all P<0.05). At 6 and 12 months after receiving HAART, plasma IL-35 was positively correlated with IL12 rβ2 and gp130, and negatively correlated with HIV-1 RNA load(all P<0.05), at the same time, gp130 was negatively correlated with HIV-1 RNA load(all P<0.05). At 12 months after receiving HAART, plasma IL12 rβ2 was negatively correlated with HIV-1 RNA load(r=-0.457, P<0.001). Conclusion The plasma levels of IL-35 and its receptors IL12 rβ2 and gp130 in HIV/AIDS patients receiving HAART increase gradually with the prolongation of HAART time, and they are closely related to HIV-1 RNA viral load. Plasma IL-35 and its receptors IL12 rβ2 and gp130 may play a role in anti HIV infection.
