Determination of the content of five components in crossbow liquid by HPLC

摘要 目的建立高效液相色谱法(HPLC)测定弩药液中八角枫碱、儿茶素、绿原酸、新乌头碱、次乌头碱5种成分的含量。方法采用色谱柱Agilent 5 TC-C18(2)(250.0 mm×4.6 mm,5μm),以乙腈-0.2%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL/min,柱温25℃、30℃,检测波长258、235 nm,进样量10μL。结果八角枫碱、儿茶素、绿原酸、新乌头碱、次乌头碱分别在2.797~14.161、25.816~130.694、7.956~40.275、2.675~13.540、1.285~6.504μg/mL的线性范围内呈现良好的线性关系(r≥0.9996);平均回收率在99.374%~101.559%,RSD在1.144%~2.844%。结论该方法快速、高效、灵敏度高,应用范围广,能为弩药液的临床疗效提供理论依据。 Objective To establish a high performance liquid chromatography(HPLC)for the determination of five components of anabasine,catechin,chlorogenic acid,mesaconitine and hypoaconitine in crossbow liquid.Methods Chromatographic column Agilent 5 TC-C18(2)(250.0 mm×4.6 mm,5μm)was used,with acetonitrile-0.2%phosphoric acid aqueous solution as the mobile phase,gradient elution,flow rate was 1.0 mL/min,column temperature was 25℃and 30℃,the detection wavelengths were 258 nm and 235 nm,and the injection volume was 10μL.Results The anabasine,catechin,chlorogenic acid,mesaconitine and hypoaconitine showed good linear relationship in the range of 2.797-14.161,25.816-130.694,7.956-40.275,2.675-13.540,1.285-6.504μg/mL respectively(r≥0.9996);the average recovery rate was between 99.374%and 101.559%,and the RSD was between 1.144%and 2.844%.Conclusion The method is fast,efficient,sensitive and has a wide range of applications.It can provide a theoretical basis for the clinical efficacy of crossbow liquid solution.
