不同降温模式下高加索三叶草的转录组比较分析 被引量:4

Comparative Analysis of Transcriptome of Caucasian Clover Trifolium ambiguum Bieb.under Different Cooling Modes

摘要 为探讨高加索三叶草(Trifolium ambiguum Bieb.)对不同降温模式低温胁迫的响应机制,本试验以缓慢降温(Gradual cooling,GC)和骤然降温(Sudden cooling,SC)处理对其进行了胁迫处理。通过转录组测序,比较了2个低温处理组与常温对照组(CK)间的转录组差异,并筛选了抗寒相关的基因和转录因子。结果表明:与CK相比,GC组和SC组中分别有8020个和6289个差异表达基因(DEGs);GC组的DEGs主要在光合作用、光合作用天线蛋白、淀粉和蔗糖代谢等通路显著富集,SC组的DEGs主要在淀粉和蔗糖代谢、倍半萜和三萜生物合成、类黄酮生物合成等通路显著富集。此外,2处理组共有的DEGs仅在淀粉和蔗糖代谢通路显著富集;一些涉及淀粉和蔗糖代谢与植物激素信号转导通路的基因在2种降温模式下均上调表达,可作为潜在的抗寒基因,如BAM3,BFRUCT1,SUS,GH3.1,SAPK2等。进一步分析发现响应低温胁迫的转录因子主要分布在AP2/ERF,ZFP,MYB等家族。 To explore the response mechanisms of Caucasian clover(Trifolium ambiguum Bieb.)to low temperature stresses under different cooling modes,gradual cooling(GC)and sudden cooling(SC)were used in this study.Through transcriptome sequencing,the transcriptome differences between two treatment groups and a control group(CK)were compared,and genes and transcription factors related to cold resistance were also screened.Compared to CK,there were 8020 and 6289 differentially expressed genes(DEGs)in GC and SC,respectively.The DEGs in GC were mainly enriched in photosynthesis,photosynthesis-antenna proteins and starch and sucrose metabolism.The DEGs in SC were mainly enriched in starch and sucrose metabolism,sesquiterpenoid and triterpenoid biosynthesis,and flavonoid biosynthesis.In addition,the DEGs shared in two treatment groups were only significantly enriched in starch and sucrose metabolism.The expression levels of some genes involved in starch and sucrose metabolism and plant hormone signal transduction pathways were up-regulated in two cooling modes.Genes such as BAM3,BFRUCT1,SUS,GH3.1,SAPK2 were identified as potential cold resistance genes.Further analysis showed the transcription factors that respond to low temperature stress were mainly distributed in AP2/ERF,ZFP,MYB and other families.
