失眠血虚证小鼠动物模型的模拟和证候评价 被引量:8

摘要 目的初步建立并评价疾病与证候相结合的失眠血虚证动物模型。方法将20只昆明雄性小鼠随机分为对照组与模型组,每组10只。模型组腹腔注射350 mg/(kg·d)对氯苯丙氨酸(p-chlorophenylalanine,PCPA),250 mg/(kg·d)环磷酰胺(cyclophosphamide,CTX)制备失眠血虚证复合动物模型,对照组注射等体积生理盐水。通过观察睡眠潜伏期、睡眠时间、昼夜节律,检测外周血,记录体表表征评分、体质量、饮水量、摄食量、胸腺指数、脾脏指数等指标对模型进行评价。结果模型组小鼠的睡眠时间显著缩短,睡眠潜伏期延长,昼夜节律消失,小鼠血虚体表表征表现为口唇、爪、甲淡白或苍白,毛发易脱落,行动倦怠、精神不振,白细胞(white blood cell,WBC)、红细胞(red blood cell,RBC)、血小板(platelet,PLT)、血红蛋白(hemoglobin,HGB)减少;胸腺指数、脾脏指数显著下降;体质量减轻、饮水量与摄食量减少,提示失眠血虚证模型成功。结论CTX+PCPA复合模型制作小鼠失眠血虚证的方法成功模拟了患者临床部分病理表现,符合失眠血虚证的证候评价,为失眠血虚证的研究提供基础。 Objective To preliminary establish and evaluate an animal model of insomnia with blood deficiency syndrome combining disease and syndrome.Methods Twenty Kunming male mice were randomly divided into a control group and a model"group,with 10 in each group.The model group was intraperitoneally injected with 350 mg/(kg·d)p-chlorophenylalanine(PCPA)and 250 mg/(kg·d)cyclophosphamide(CTX)to prepare a composite animal model of insomnia with blood deficiency pattern,and the control group was injected with equal volume of saline.The model was evaluated by observing sleep latency,sleep time,circadian rhythm,detecting peripheral blood,recording body surface characterization score,body mass,water intake,food intake,thymus index and spleen index.Results The sleep time of mice in the model group was significantly shortened,the sleep latency was extended,and the circadian rhythm disappeared,and the symptoms of blood deficiency appeared such as pale lips,paws,and nails,loss of hair,fatigue,lassitude,decrease of white blood cells(WBC),red blood cells(RBC),platelets(PLT),hemoglobin(HGB),and significant decrease of thymus index and spleen index,decreased weight,water intake and food intake,which suggested the success of the insomnia blood deficiency syndrome model.Conclusions The method of CTX+PCPA compound model to make mice with insomnia and blood deficiency syndrome successfully simulated part of the patient’s clinical pathology,which is in line with the evaluation of insomnia with blood deficiency syndrome and provides a basis for future research on insomnia mechanisms.
