基于Wnt/β-catenin通路研究麝香保心丸对冠心病心力衰竭大鼠的作用 被引量:7

Effects of Shexiang Baoxin Pills on Rats With Coronary Heart Disease and Heart Failure Based on Wnt/β-Catenin Signaling Pathway

摘要 目的:探究麝香保心丸对冠状动脉粥样硬化性心脏病心力衰竭大鼠心肌损伤、心肌细胞氧化应激及对Wnt/β-catenin通路的影响。方法:将大鼠随机分为假手术组、模型组、卡托普利(135 mg·kg^(-1))组及麝香保心丸组(低、中、高剂量)(25 mg·kg^(-1)、50 mg·kg^(-1)、75 mg·kg^(-1))组,除假手术组,其余大鼠复制冠心病心力衰竭模型后,给予相应的药物灌胃4周。检测大鼠左心室舒张末期内径(left ventricular end-diastolic dimension,LVDd)、左心室收缩末期内径(left ventricular end-systolic dimension,LVDs)、左心室舒张末期容积(left ventricular end-diastolic volume,LVEDV)、左心室收缩末期容积(left ventricular end-systolic volume,LVESV)、左室质量指数(left ventricular mass index,LVMI)、体质量(body mass,BM)、心脏质量(heart mass,HM)、全心重指数(total heart mass index,HWI);检测血浆脑钠肽(brain natriuretic peptide,BNP)水平;检测心肌组织凋亡指数(apoptosis index,AI)、活性氧(reactive oxygen species,ROS)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活性;检测心肌组织β-catenin mRNA、GSK-3βmRNA及LRP6 mRNA水平。结果:与模型组比较,麝香保心丸低、中、高剂量组及卡托普利组大鼠LVDs、LVDd、LVESV、LVEDV、HWI、LVMI、AI均降低,血浆BNP水平显著降低,心肌组织ROS、MDA活力及LRP6 mRNA、GSK-3βmRNA、3β-catenin mRNA水平均显著降低。结论:麝香保心丸可减轻冠状动脉粥样硬化性心脏病心力衰竭大鼠心室重构,抑制心肌组织氧化应激反应,其作用机制可能与抑制Wnt/β-catenin通路有关。 Objective:To explore the effects of Shexiang Baoxin Pill on myocardial injury,myocardial oxidative stress,and Wnt/β-catenin signaling pathway in rats with coronary heart disease(CHD)with heart failure.Methods:The rats were randomly divided into sham operation group,model group,captopril(135 mg·kg^(-1))group,and Shexiang Baoxin pill group(low,medium and high dose)(25 mg·kg^(-1),50 mg·kg^(-1),75 mg·kg^(-1))groups,except for the sham operation group,the rest of the rats were given the corresponding drugs by gavage for 4 weeks after replicating the CHD heart failure model.After the treatment,the left ventricular end-diastolic dimension(LVDd),left ventricular end-systolic dimension(LVDs),left ventricular end-diastolic volume(LVEDV),Left ventricular end-systolic volume(LVESV),left ventricular mass index(LVMI),body mass(BM),heart mass HM),total heart mass index(HWI)was measured.The levels of plasma brain natriuretic peptide(BNP),myocardial tissue apoptosis index(AI),reactive oxygen species(ROS),superoxide dismutase(SOD),malondialdehyde(MDA),and glutathione peroxidase(GSH-Px)activity were detected.And myocardial tissueβ-catenin mRNA,GSK-3βmRNA,and LRP6 mRNA levels were detected.Results:Compared with the model group,LVDs,LVDd,LVESV,LVEDV,HWI,LVMI,AI in the low,medium,and high dose groups of Shexiang Baoxin pill groups and captopril group were all decreased,the plasma BNP level was significantly decreased,ROS,MDA activity,and LRP6 mRNA,GSK-3βmRNA,3β-catenin mRNA levels in myocardial tissue were significantly reduced.Conclusion:Shexiang Baoxin Pill can alleviate ventricular remodeling and inhibit the oxidative stress response of myocardium in CHD rats with heart failure.Its mechanism may relate to the inhibition of the Wnt/β-catenin signaling pathway.
