摘要
枯草芽孢杆菌是一种有益的微生物,在土壤、空气、水和腐败的有机物中都有分布,有消毒抑菌防腐、水生态保护、除臭、提高免疫力等功能,可应用在生态发酵、微生物医学、动植物的生产和保护、粪污环境污染控制等方面。文章对枯草芽孢杆菌的成熟枯草杆菌蛋白酶基因进行分析发现,密码子第3位以A、T为主,密码子偏性中等,可利用二步重叠PCR法等进一步优化表达;有较多的优势密码子,表达水平提升空间大,可进一步选择大肠杆菌、酵母、芽孢杆菌、昆虫或者哺乳动物等比较一致的表达载体系统转化表达。
Bacillus subtilis is a beneficial microorganism,which is distributed in soil,air,water and putrefied organic matter.It has the functions of disinfection,bacteriostasis,antisepsis,water ecological protection,deodorization,immunity improvement and can be applied to the improvement of ecological fermentation,microbial medicine,production and protection of animals and plants,and environmental pollution control of manure pollution.It is of great significance for practical application to study the codon of Bacillus subtilis protease gene.In this paper,the mature subtilisin gene of Bacillus subtilis was analyzed,and it was found that the third codon position was mainly A and T,and the codon bias was moderate,which could be further optimized by two-step overlapping PCR There are more dominant codons,and the expression level has a large space for improvement,so we can further select the expression vector system which is consistent with Escherichia coli,yeast,Bacillus,insects or mammals for transformation and expression.
