沙参麦冬汤的HPLC指纹图谱研究 被引量:4

摘要 目的建立经典名方沙参麦冬汤的HPLC指纹图谱,同时结合聚类分析及主成分分析对其进行评价,为沙参麦冬汤的质量控制提供科学依据。方法采用Zorbax C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.2%磷酸水为流动相,梯度洗脱,建立15批次不同产地的沙参麦冬汤的HPLC指纹图谱,并采用《中药色谱指纹图谱相似度评价系统2012A版》及SPSS 21.0软件对15批沙参麦冬汤的HPLC指纹图谱进行相似度评价、聚类分析及主成分分析。结果建立了15批沙参麦冬汤的HPLC指纹图谱,相似度均>0.90,标定了26个共有峰,指认出3个色谱峰,分别为13号峰(芦丁)、15号峰(甘草苷)及26号峰(甘草酸铵);聚类分析验证了相似度分析的结果;主成分分析揭示了指纹图谱中对整体信息贡献较大的特征峰。结论所用方法快速、简便、重复性好,可为沙参麦冬汤及其制剂的质量评价提供参考。 OBJECTIVE To establish an HPLC fingerprint of Shashen Maidong decoction(SMD)combined with cluster analysis and principal component analysis,and to provide scientific basis for the quality control of SMD.METHODS The HPLC fingerprints of 15 batches of SMD from different places were established by using Zorbax C18 column(250 mm×4.6 mm,5μm)and acetonitrile-0.2%phosphoric acid water as mobile phase.The similarity evaluation was carried out by using traditional Chinese medicine chromatographic fingerprint similarity evaluation system 2012A,and the cluster analysis and principal component analysis were carried out by using SPSS 21.0 software.RESULTS The HPLC fingerprints of 15 batches of SMD were established.The similarity was all more than 0.90,26 common peaks were calibrated and 3 chromatographic peaks were identified,which were 13(rutin),15(glycyrrhizin)and 26(ammonium glycyrrhizate),respectively.Cluster analysis verified the results of similarity analysis and principal component analysis revealed that the characteristic peaks contributed more to the overall information in the fingerprint.CONCLUSION The method is rapid,simple and reproducible,and can provide reference for the quality evaluation of SMD and its preparations.
