摘要
目的以肉苁蓉总苷为研究对象进行全成分表征分析和裂解规律研究,为后续揭示肉苁蓉总苷体内作用过程和雌激素作用质量标志物奠定基础。方法采用超高效液相色谱-四极杆飞行时间质谱技术分析肉苁蓉总苷,数据经Agilent Masshunter Qualitative Analysis B.7软件处理,将各色谱峰在不同模式下的分子离子峰及其对应的碎片离子与对照品及文献信息进行比对。结果以负离子模式为主、正离子模式为辅进行检测分析,共推测出40个化学成分,包括20个苯乙醇苷类成分、10个环烯醚萜苷类成分、4个萜类成分和6个其他成分。结合质谱碎片等信息对不同结构类型肉苁蓉总苷的裂解规律进行归纳总结,推测其主要的裂解方式是丢失葡萄糖、咖啡酰基、羟基酪醇、H_(2)O等母环上的功能基团,及糖断裂后产生的CH_(2)O、C_(2)H_(4)O_(2)等碎片。结论UPLC-Q-TOF-MS技术鉴定肉苁蓉总苷中化学成分,信息全面准确,操作高效便捷,是下一步肉苁蓉总苷入血成分分析和雌激素作用质量标志物筛选的可靠数据支持。
OBJECTIVE To reveal the quality markers of estrogenic effect and in vivo action process,analyze the chemical constituents of Cistanche deserticola total glycosides,and summarize the fragmentation patterns of different structural types.METHODS UPLC-Q-TOF-MS was used to qualitatively analyze the chemical constituents of Cistanche deserticola total glycosides.Agilent Masshunter Qualitative Analysis B.7 was used to process experimental data.The structures of chemical constituents were identified by fragment analysis,standards comparison and references.RESULTS A total of 40 chemical components of Cistanche deserticola total glycosides were inferred in the positive and negative ion scanning modes.It includes 20 phenylethanoid glycosides,10 iridoid glycosides,4 terpenoids and 6 others.Based on the mass spectrometry fragments and other information,the fragmentation patterns of different structural types of Cistanche deserticola total glycosides were summarized.It was found that the main fragmentation patterns were loss of functional groups on the mother rings of glucose,caffeoyl,hydroxytyrosol,H_(2)O,etc,and CH_(2)O,C_(2)H_(4)O_(2),etc,fragments produced after sugar breakage.CONCLUSION UPLC-Q-TOF-MS is used to identify the chemical components in Cistanche deserticola total glycosides.The experimental result is comprehensive and accurate,and the operation is efficient and convenient.It is reliable data support for analysis of absorbed components in rat plasma after oral administration of Cistanche deserticola total glycosides and screening of quality markers of estrogenic effect.
