摘要
目的探讨载脂蛋白B(APOB)在肝癌中的表达水平,并分析其在肝癌诊断和预后中的预测价值。方法通过癌症基因图谱(TCGA)、Oncomine数据库分析APOB mRNA在肝癌组织和癌旁组织中的表达;利用Kaplan Meier Plotter数据中的肝癌数据集进行在线生存分析;采用cBioportal分析工具对APOB的基因改变频率进行分析;对比肝癌患者和健康体检者两组血清APOB水平差异,分析肝癌患者血清APOB水平与临床病理参数和预后的相关性。结果APOB mRNA在肝癌组织中的表达水平低于癌旁组织(P<0.05);肝癌患者APOB蛋白水平低于健康体检者(P<0.05);APOB mRNA的表达水平低的患者总生存期(P<0.0001)和无复发生存期(P=0.0076)均短于表达水平高的患者,APOB基因突变患者的预后较无突变患者差,其表达与肿瘤分级及生存预后有关。结论APOB在肝癌组织和肝癌患者血清中表达降低,APOB表达降低与肿瘤分级和不良预后有关。
Objective To investigate the prognostic significance of the expression of Apolipoprotein B(APOB) in liver cancer by bioinformatics data and immunohistochemistry.Methods Expression of APOB mRNA in liver cancer tissues and normal adjacent tissues was investigated using data from the Cancer Genome Atlas(TCGA), and Oncomine database. Kaplan Meier Plotter identified the association between APOB mRNA and prognosis. Correlation between APOB genetic mutation and prognosis used the cBioportal for Cancer Genomics and the Mutation website. The serum level of APOB in liver cancer patients and healthy people was compared, the value of prediction efficacy and prognosis was evaluated. Results APOB mRNA was significantly lower in liver cancer tissues compared with normal tissues(P<0.05) APOB protein was significantly lower in liver cancer patients compared with healthy people(P<0.05). The results from GSE15459 showed that patients with low levels of APOB mRNA had a reduced overall survival(P<0.0001) and disease-free survival(P=0.0076). A negative correlation between APOB mRNA and DNA mutation was found using the cBioportal. Cox regression analysis showed that APOB expression was significantly correlated with tumor grade and prognosis.Conclusion Downregulation of APOB predicts poor tumor grade and prognosis in liver cancer. APOB may act as a tumor suppressor in liver cancer.
