京尼平苷酸的大鼠血浆药代动力学研究

摘要 目的:通过测定大鼠口服京尼平苷酸后血药浓度并计算药物代谢动力学参数,研究该药物的代谢情况。方法:使用SD大鼠,口服20mg/kg的京尼平苷酸药液后,在24小时内按规定时间点取血,血浆加乙腈除蛋白,使用UPLC-MS法测定京尼平苷酸的血药浓度。质谱参数为,ESI离子源,正负离子同时扫描模式,吹扫电压3000V,雾化温度350℃,毛细管温度120℃,氮气流速50L/h,监测离子对为京尼平苷酸372.8/149.1,柚皮苷(内标)579.0/300.6,碰撞电压分别为30V和25V,液相色谱条件为,使用Phenomenex Kinex C18色谱柱(4.6×50mm,5m),以0.1%甲酸(A)-乙睛(B)为流动相,梯度洗脱,梯度程序为,0-0.5min,5-5%B;0.5-1.0min,5-95%B;1.0-2.5min,95-95%B;2.5-3.0min,95-5%B;流速300L/min。结果:6只大鼠京尼平苷酸的药代动力学参数为:Cmax 629.76ng/mL,Tmax=1.50h,T1/2=1.08h,MRT=3.13h,AUC0-=1657.82h·ng/mL,AUC0-∞=1677.35 h·ng/mL。结论:本研究所用方法快速、准确、高效,能够较好地评价京尼平苷酸在体内的运动过程。 Objective:to investigate the absorption,distribution,metabolism and elimination of geniposidic acid by determination of its plasma contents and calculation of pharmacokinetic parameters after oral administration by rats.Methods:blood was taken from SD rats at set intervals within24 h after orally taking 20 mg/kg of geniposidic acid;when removing proteins by adding acetonitrile,the plasma concentration of geniposidic acid was analyzed by UPLC-MS.The Mass spectrometry parameters were:ESI ion source,positive and negative scan mode,spray voltage was 3000 V,vaporized temperature was 350℃,and the capillary temperature was 120℃,the nitrogen flow rate was 50 L/h,and the ion pairs for geniposidic acid was372.8/149.1,for Naringin(the internal standard) was 579.0/300.6,the collision energy was 30 V and 25 V,respectively;the liquid chromatography condition was,an Phenomenex Kinex C18 column(4.6×50 mm,5 μm) was used,the mobile phase was 0.1% formic acid-acetonitrile,with the following gradient program:0-0.5 min,5-5% B;0.5-1.0 min,5-95%B;1.0-2.5 min,95-95%B;2.5-3.0 min,95-5%B;the flow rate was 300μL/min;6 rats were analyzed.Results:in 6 rats,the pharmacokinetic parameters were:Cmax 629.76 ng/mL,Tmax=1.50 h,T1/2=1.08 h,MRT=3.13 h,AUC0-t=1657.82 h·ng/mL,AUC0-∞=1677.35 h·ng/mL.Conclusion:the proposed method was fast,accurate and efficient,which can be used to evaluate in body dynamics of geniposidic acid.
