ZmCDPK7增强玉米原生质体的耐热性分析 被引量:3

Analysis on ZmCDPK7 enhancing the thermotolerance of maize protoplasts

摘要 以玉米品种郑单958为试验材料,对玉米中40个ZmCDPKs的主要结构进行生物信息学技术分析,显示它们的结构域具有很高的保守性。利用qRT-PCR对这40个ZmCDPKs表达进行分析,发现ZmCDPK7表达显著受高温诱导。通过在玉米叶原生质体瞬时过表达或沉默ZmCDPK7来分析其作用,结果显示,与无高温胁迫相比,瞬时过表达ZmCDPK7显著促进了高温胁迫诱导的小热休克蛋白sHSP17.4、热休克蛋白HSP70、过氧化氢酶CAT1、抗坏血酸过氧化物酶APX1、呼吸爆发氧化酶同源物RBOHA和RBOHB的表达及CAT和APX的活性增加,但降低了高温胁迫诱导的MDA,H_(2)O_(2)含量增加;而瞬时沉默ZmCDPK7的作用相反。 In this study,maize variety Zhengdan 958 was used as the experimental material.The bioinformatic analysis of 40 ZmCDPKs structure in maize showed that the domains of these 40 ZmCDPKs were highly conservative.The expression of 40 ZmCDPKs was analyzed by qRT-PCR,and the results indicated that the expression of ZmCDPK7 induced by high temperature.The roles of ZmCDPK7 were analyzed by its transient overexpression or RNAi silence in maize protoplasts.The results showed that compared with no high temperture stress,ZmCDPK7 transient overexpression significantly promoted heat-induced gene expression of small heat shock protein-sHSP17.4,heat shock protein-HSP70,catalase 1-CAT1 and ascorbate peroxidase 1-APX1,respiratory burst oxidase homologues RBOHA and RBOHB,and the enzyme activity of CAT and APX,but decreased the heat-induced MDA and H_(2)O_(2) content.Whereas,the transient silencing of ZmCDPK7 showed the opposite results.
