摘要
目的考察安神益智合剂对力竭游泳小鼠的抗疲劳作用及机制。方法采用一次性力竭游泳方式制作运动性小鼠疲劳模型,记录小鼠力竭游泳时间,用分光光度法测定血清尿素氮(BUN)、乳酸(LD)、乳酸脱氢酶(LDH)、肌糖原和肝糖原含量,酶联免疫法测定睾酮和皮质酮含量,RT-PCR和Western Blot法测定肌肉组织中相关基因的转录和翻译水平。结果与正常组比较,模型组小鼠血清睾酮/皮质酮(T/C)比值、肝糖原和肌糖原含量均显著下降,而血清LD、LDH和BUN含量均显著升高。与模型组比较,安神益智合剂给药组小鼠的力竭游泳时间显著延长,血清T/C比值、肝糖原和肌糖原含量均升高,血清LD、LDH和BUN含量均降低;小鼠肌肉组织中CYP17A1和CYP11A1基因转录和翻译水平均显著升高。结论安神益智合剂能促进力竭小鼠肌肉组织中CYP17A1和CYP11A1 mRNA及蛋白表达水平,从而调节睾酮、皮质酮水平,提高T/C比值,促进蛋白质合成,达到抗疲劳的作用。
Objective To investigate the anti-fatigue effect and mechanism of Anshen Yizhi Mixture in exhausted swimming mice.Methods A one-time exhaustive swimming method was used to make an mouse fatigue model,and the time of exhausted swimming in mice was recorded.The serum urea nitrogen(BUN),lactic acid(LD),lactate dehydrogenase(LDH),and liver and muscle glycogen content were determined by spectrophotometry.The content of testosterone and corticosterone was measured by enzyme-linked immunoassay.The transcription and translation levels of related genes in muscle tissue were measured by RT-PCR and Western Blot.Results Compared with the normal group,the serum testosterone/corticosterone(T/C)ratio,liver and muscle glycogen contents of the model group were significantly decreased,while the serum LD,LDH and BUN contents were significantly increased.Compared with the model group,the mice in the Anshen Yizhi Mixture administration group had significantly longer exhaustive swimming time,and the serum T/C ratio,liver and muscle glycogen contents were increased.The serum contents of LD,LDH and BUN were reduced,the CYP17A1 and CYP11A1 gene transcription and translation levels in mouse muscle tissue were significantly increased.Conclusion Anshen Yizhi Mixture may promote CYP17A1 and CYP11A1 mRNA and protein expression levels in the muscle tissues of exhausted mice,thereby regulate the levels of testosterone and corticosterone,increase the T/C ratio,promote the protein synthesis,and achieve the anti-fatigue effects.
