五味子复合提取物对高脂血症大鼠的血脂调节作用及其机制 被引量:8

Regulatory Effect of Schisandra Compound Extract on Blood Lipids in Hyperlipidemic Rats and Its Mechanism

摘要 目的探讨五味子复合提取物对高脂血症大鼠血脂水平的调节作用及作用机制.方法将32只Wistar雄性大鼠适应性喂养1周后,随机分为空白对照组、模型组、低高剂量五味子复合提取物组,每组8只.除空白对照组外,其余3组均采用高脂饲料喂养诱导高脂血症大鼠模型.低、高剂量五味子复合提取物组大鼠分别灌胃给予100、200 mg/kg五味子复合提取物,空白对照组和模型组灌胃给予等体积的蒸馏水,共给药8周.检测各组大鼠血清中甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)水平,肝脏TC、TG水平.苏木素-伊红(HE)染色观察各组大鼠肝脏病理学变化,RT-PCR和Western blotting法检测各组大鼠肝脏组织中PPARα、LXRα、CYP7A1、SREBP2和HMGCR的表达水平.结果与空白对照组比较,模型组大鼠体质量、肝脏指数、血清TC、TG、LDL-C水平均明显增加(P<0.01)、HDL-C水平明显降低(P<0.01),肝中TC、TG水平明显增加(P<0.01或P<0.05),肝脏组织中产生脂质沉积,PPARα、LXRα、CYP7A1表达水平明显降低(P<0.01),SREBP2、HMGCR表达水平明显升高(P<0.01).与模型组比较,低、高剂量五味子复合提取物组大鼠的体质量、肝指数、血清中TC、TG、LDL-C水平均明显降低(P<0.05或P<0.01),HDL-C水平明显升高(P<0.05),肝组织中TC、TG水平明显降低(P<0.01或P<0.05),肝脏脂质沉积减轻,PPARα、LXRα、CYP7A1表达水平明显增加(P<0.01或P<0.05),SREBP2、HMGCR表达水平明显降低(P<0.01或P<0.05).与低剂量五味子复合提取物组比较,高剂量五味子复合提取物组血清中TC、TG、LDL-C水平明显降低(P<0.05),HDL-C水平明显升高(P<0.05),肝中TC水平明显降低(P<0.05),PPARαmRNA表达明显增加(P<0.05),SREBP2、HMGCR mRNA表达明显降低(P<0.05).结论五味子复合提取物对高脂血症大鼠的血脂具有一定的调节作用,其机制可能与其调节PPARα/LXRα/CYP7A1和SREBP2/HMGCR信号通路的表达有关. Objective To study the lipid-regulating effect of Schisandra compound extract(SCE)on hyperlipidemic rats and its mechanism.Method After 32 male Wistar rats were adaptively fed for 1 week,they were randomly divided into control group,model group,low and high dose of SCE groups,with 8 rats in each group.Except the control group,rats in the other 3 groups were fed with a high-fat diet to induce the rat hyperlipidemic model.The rats in low and high doses of SCE groups were administered with 100,200 mg/kg of Schisandra compound extract by gavage,respectively.The rats in control group and model group were intragastrically given the same volume of distilled water once daily for 8 weeks.The levels of triglyceride(TG),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C)in serum of rats,the TG and TC contents in the liver tissues were measured.HE staining was performed for observing histopathological changes in liver.RT-PCR and Western blotting were used to detect the expressions of PPARα,LXRα,CYP7A1,SREBP2 and HMGCR in the liver of rats.Results Compared with control group,the body weight,the liver index,serum TC,TG,LDL-C levels,TC and TG contents in the liver tissues of model groups significantly increased(P<0.01),and the level of serum HDL-C significantly decreased(P<0.01),lipid deposition in the liver of rats was occurred,the expression levels of PPARα,LXRαand CYP7A1 significantly decreased(P<0.01),the expression levels of SREBP2 and HMGCR significantly increased(P<0.01).Compared with model group,the body weight,the liver index,serum TC,TG,LDL-C levels of low and high doses of SCE groups significantly decreased(P<0.05 or P<0.01),and the level of serum HDL-C significantly increased(P<0.05),TC and TG contents in the liver tissues significantly decreased(P<0.01 or P<0.05),lipid deposition in the liver of rats reduced,the expression levels of PPARα,LXRαand CYP7A1 significantly increased(P<0.01 or P<0.05),the expression levels of SREBP2 and HMGCR significantly decreased(P<0.01 or P<0.05).Compared with low doses of SCE group,serum TC,TG,LDL-C levels of high doses of SCE group significantly decreased(P<0.05),and the level of serum HDL-C significantly increased(P<0.05),TC content in the liver tissues significantly decreased(P<0.05),the expression level of PPARαmRNA significantly increased(P<0.05),the expression levels of SREBP2,HMGCR and mRNA significantly decreased(P<0.05).Conclusion Schisandra compound extract has a certain regulatory effect on blood lipids in hyperlipidemic rats,which may be related to its regulation of the expression of PPARα/LXRα/CYP7A1 and SREBP2/HMGCR signaling pathways.
