发酵中草药对蛋鸡产蛋后期肝脏mRNA表达的影响 被引量:2

Effects of Fermented Chinese Herbal Medicines on Liver mRNA Expression of Laying Hens in Late Laying Stage

摘要 探究发酵中草药(主要成分为党参、白术、白芍和茯苓,发酵微生物为枯草芽孢杆菌)调控蛋鸡产蛋后期肝脏与脂代谢相关基因的分子机制,为改善蛋鸡的生产性能打下基础。试验选取体重相近、饲养管理水平一致,并处于产蛋后期的京红蛋鸡(52周龄)480只,随机分为2组,每组4个重复,每个重复60只鸡。对照组饲喂基础饲粮,试验组在基础饲粮中添加4 g/kg发酵中草药。结果表明:1)在产蛋性能方面,相较于对照组,试验组产蛋率显著提升了7.59%(P<0.05),料蛋比极显著降低了0.97%(P<0.01)。2)对试验组和对照组进行差异表达分析后,共筛选出1 039个差异表达基因(上调642个,下调397个)。基因本体论(GO)分析显示,差异表达基因显著富集在脂肪酸分解代谢过程、细胞脂质分解代谢过程及脂质分解代谢过程等通路;京都基因和基因组百科全书(KEGG)通路分析显示,差异表达基因显著富集到脂肪酸代谢、过氧化物酶体增殖物激活受体(PPAR)信号通路、脂肪酸降解及脂肪细胞因子信号通路等途径。综上所述,本试验在添加发酵中草药后可有效提高京红蛋鸡产蛋后期产蛋率,并利用转录组测序技术,初步揭示了鸡肝脏脂代谢调控的分子机制。 In this study,transcriptome sequencing(RNA-seq) technology was used to explore the molecular mechanism of fermented Chinese herbal medicines(The main components of fermented Chinese herbal medicines are Codonopsis,Atractylodes,Radix Paeoniae Rubra and Poria,and the fermenting microorganism is Bacillus subtilis)that would regulate liver lipid metabolism in laying hens.In the experiment,480 Jinghong laying hens(52 weeks old) with similar body weights and consistent feeding in the late laying stage and management levels were selected. After that they were randomly divided into 2 groups with 4 replicates in each group and 60 chickens in each replicate. The control group was fed basal diet,and the experimental group was fed with 4 g/kg fermented Chinese herbal medicines in the basal diet.The results showed that:1) in terms of egg production performance,compared with the control group,the egg production rate of the experimental group was significantly increased by7.59%(P<0.05),and the feed-to-egg ratio was extremely significantly reduced by 0.97%(P<0.01). 2) After the differential expression analysis of the experimental group and the control group,a total of 1039 differentially expressed genes(642 up-regulated and 397 down-regulated) were screened out. Gene ontology(GO) analysis showed that differentially expressed genes were significantly enriched in fatty acid catabolism,cellular lipid catabolism,and lipid catabolism pathways;The Kyoto Encyclopedia of Genes and Genomics(KEGG) pathway analysis showed that differentially expressed genes were significantly enriched in fatty acid metabolism,peroxisome proliferator-activated receptor(PPAR) signaling pathway,fatty acid degradation,and adipocytokine signaling pathway,etc. In summary,this experiment can effectively increase the egg production rate of Jinghong laying hens after fermented Chinese herbal medicines are added,and the molecular mechanism of lipid metabolism regulation in chicken liver is revealed by transcriptome sequencing technology.
