摘要
基于四通阀在线稀释装置和单通道雾化室,建立了在线稀释进样-电感耦合等离子体质谱(ICP-MS)测定单个酵母细胞中痕量元素的分析方法。结果表明,在线稀释装置与单通道雾化室联用的单细胞信号采集效率为31.3%,显著大于双通道雾化室的1.3%。所测富硒酵母细胞中元素信号呈现不同的分布情况,除Mg外,P、Cu、Zn、Se等都表现为明显的多峰分布,提示这些元素含量与酵母细胞大小及其生理状态之间可能存在关联。本方法设计简单、易于操作,可用于向ICP-MS仪中直接引入含一定浓度缓冲盐的细胞悬液,在单细胞元素分析领域具有较好的实际应用价值。
A new method for the determination of elements in single cells by on-line dilution sampling inductively coupled plasma mass spectrometry(ICP-MS)was developed,based on a on-line dilution device of the four-way valve and a single-channel spray chamber,and applied to the measurement of inorganic elements in single yeast cells.The results show that the single-cell signal acquisition efficiency of the on-line dilution device combined with the single-channel chamber is 31.3%,which is significantly higher than that of the double-channel chamber of 1.3%.Except for Mg,P,Cu,Zn and Se all show obvious multi peak distribution,suggesting that there may be a relationship between the content of these elements and the size and physiological state of yeast cells.The proposed method is simple in design and easy to operate.It can be used to directly introduce cell suspension samples with certain concentration of buffer salt into ICP-MS,so it will have good applications in the field of single-cell element analysis.
