草苁蓉多糖提取物对喉癌细胞增殖、迁移及程序性死亡分子1/程序性死亡配体-1水平的研究分析 被引量:7

Effect of Cistanche deserticola extract on proliferation,migration and PD-1P/D-L1 level of laryngeal carcinoma cells

摘要 目的探究草苁蓉多糖提取物(BRP)对喉癌细胞增殖、迁移及程序性死亡分子1(programmed death-1 receptor,PD-1)/程序性死亡配体-1(programmed cell death-ligand 1,PD-L1)水平的影响。方法人喉癌Hep-2细胞分组,根据BRP浓度分为YY组(100 mg/L)、EY组(200 mg/L)、SY组(400 mg/L)及WY组(无药物干扰)。采用qRT-PCR、克隆、MTT法、Transwell小室及Western blot检测检测4组Hep-2细胞中的PD-1/PD-L1 mRNA表达、细胞复制、细胞活力、迁移能力及PD-1/PD-L1蛋白变化。结果WY组Hep-2细胞表现为船形,生长较快,细胞分布密集。YY组细胞生长速度减慢,EY组/SY组细胞体积缩小、固缩及碎裂。YY组、EY组、SY组Hep-2细胞抑制率升高,克隆细胞及迁移数量均降低,相比WY组有差异(P均<0.05);相比WY组,YY组、EY组、SY组Hep-2细胞中的PD-1/PD-L1表达均降低(P均<0.05)。结论BRP能够通过降低PD-1/PD-L1表达抑制Hep-2细胞增殖和迁移,并且药效的发挥随时间和浓度增加而增大。 OBJECTIVE To investigate the effects of BRP extract on the proliferation,migration and PD-1/PD-L1 level of laryngeal carcinoma cells.METHODS Human laryngeal cancer cells Hep-2 were divided into YY group(100 mg/L),EY group(200 mg/L),SY group(400 mg/L)and WY group(without drug interference)according to BRP concentration.QRT-PCR,cloning,MTT assay,Transwell chamber and Western blot were used to detect PD-1/PD-L1 mRNA expression,cell replication,cell viability,migration ability and PD-1PD-L1 protein changes in Hep-2 cells of the four groups.RESULTS Hep-2 cells in WY group showed a boat like appearance with rapid growth and dense distribution.The growth rate of the cells in the YY group was slowed down,while the volume of the cells in the EY/SY group was reduced,pyropysis and fragmentation.The inhibitory rate of Hep-2 cells in YY group,EY group and SY group was increased,and the number of clone cells and migration were decreased,compared with that in WY group(P<0.05).Compared with WY group,the expression of PD-1/PD-L1 in Hep-2 cells in YY group,EY group and SY group was decreased(P<0.05).CONCLUSION Cistanche deserticola extract can inhibit the proliferation and migration of laryngeal cancer cell line Hep-2 by reducing the expression of PD-1/PD-L1,and the efficacy increases with the increase of time and concentration.
