摘要
目的:通过网络药理学方法探讨赤芍⁃牡丹皮药对治疗异常子宫出血的作用机制。方法:检索中药系统药理学平台选取赤芍⁃牡丹皮药对的有效元素和作用靶点;通过检索人类基因信息数据库选取异常子宫出血的疾病靶点;应用R语言筛选药物与疾病共同靶点,应用Cytoscape软件构建药物⁃化合物⁃作用靶点⁃疾病互作网络;运用STRING平台构建蛋白质间相互作用网络(PPI),并将结果进行可视化展示;使用Bioconductor生物信息软件包进行GO功能富集分析和KEGG通路分析。结果:本次研究包含了16个化合物和67个关键作用靶点。富集分析得到87个GO功能条目和116条KEGG信号通路。赤芍⁃牡丹皮药对中的槲皮素、黄芩素、山柰酚、豆甾醇等成分直接作用于白细胞介素⁃6、表皮生长因子受体、胱天蛋白酶3、丝裂原激活蛋白激酶8和血管内皮生长因子等相关靶点起作用,并主要富集在Kaposi sarcoma⁃associated herpesvirus infection、Hepatitis B、AGE⁃RAGE、TNF等多条信号通路上。结论:赤芍⁃牡丹皮药对可能通过多成分多靶点发挥抗细胞增殖和凋亡、保护血管内皮、抗炎症反应、调节激素分泌、提高抗氧化活性等作用,起到治疗异常子宫出血的目的。
Objective:To explore the mechanism of the treatment of abnormal uterine bleeding with Radix Paeoniae Ru⁃bra⁃Cortex Moutan.Methods:The effective elements and action targets of Radix Paeoniae Rubra⁃Cortex Moutan were searched by the pharmacology platform of traditional Chinese medicine system,and the disease targets of abnormal uterine bleeding selected by searching the human gene information database.R language was used to screen the common targets of drugs and diseases.More⁃over,the interaction network of drugs⁃compounds⁃action targets⁃diseases was constructed through the Cytoscape software and the protein⁃protein interaction network was established through the string platform.The Bioconductor bioinformation software package was used to perform GO function enrichment and KEGG pathway analyses.Results:This study included 16 compounds and 67 key targets.After enrichment analyses,87 GO functional items and 116 KEGG signaling pathways were obtained.The constitu⁃ents such as quercetin,scutellarin,kaempferol and stigmasterol from Radix Paeoniae Rubra⁃Cortex Moutan directly act on the tar⁃gets including interleukin⁃6,epidermal growth factor receptor,caspase 3,mitogen activated protein kinase 8,vascular acted endothelial growth factor,and were mainly enriched in Kaposi sarcoma associated herpesvirus infection,hepatis B,AGE⁃RAGE,TNF and other signaling pathways.Conclusion:Radix Paeoniae Rubra⁃Cortex Moutan may play a key role in inhibition of cell pro⁃liferation and apoptosis,protection of vascular endothelium,anti⁃inflammatory response,regulation of hormone secretion,and im⁃provement of antioxidant activity through multiple components and targets in the treatment of abnormal uterine bleeding.
