摘要
为选育出在35℃~38℃能正常生长的灵芝菌株,采用紫外诱变法将制备好的灵芝原生质体置于紫外灯下距离20 cm照射30 s进行诱变,诱变后的原生质体经过5 d、28℃培养后,再置于51℃下热激2 h,观察灵芝菌丝的恢复生长情况以筛选耐高温菌株。结果表明,经过多次筛选与纯化,筛选出耐高温灵芝菌株10株。其中37℃下编号为Y1和Y30的诱变菌株生长情况最好。通过紫外诱变原生质体的方法,筛选出了能在37℃高温下生长的灵芝新菌株,为灵芝的基础研究及其他耐高温菌种的选育提供了一定的参考。
In order to screen Ganoderma lucidum strains that can grow normally at a temperature range from 35℃ to 38℃, the prepared protoplasts of G. lucidum were irradiated by ultraviolet lamp at a distance of 20 cm for 30 s using UV mutagenesis.The irradiated protoplasts were heat shocked at 50℃ for 2 h after cultureing for 5 days at 28℃, and the recovery growth of G.lucidum mycelium were observed to screen the high temperature resistant strains. The results showed that 10 strains of high temperature resistant G. lucidum strains were screened after repeated screening and purification. Among them, the mutagenized strains numbered Y1 and Y30 had the best growth at 37℃. New strains of G. lucidum that can grow at 37℃ were screened by ultraviolet mutagenesis of protoplasts, which provides a certain reference value for the basic research of G. lucidum and the breeding of other high temperature resistant strains.
