摘要
以红果肉苹果‘红脆甜’二倍体及其同源四倍体植株为材料,利用临时装片和石蜡切片比较两种倍性植株叶片显微结构差异,测定表皮细胞和气孔器密度,测量栅栏组织、海绵组织以及上下表皮厚度,分析加倍后叶片结构的变异特征。结果表明,四倍体植株比之二倍体,叶片的表皮细胞密度和气孔器密度发生了极显著变化,表皮细胞和气孔器同步变大,使得单位视野的密度明显变小;四倍体叶片的上下表皮、栅栏组织、海绵组织均较二倍体明显增厚。
Using the diploid and autotetraploid plants of red flesh apple ‘Hongcuitian’ as materials, the differences of leaf microstructure were compared by using temporary film and paraffin section. The density of epidermal cells and stomatal apparatus were measured, the thickness of palisade tissue, spongy tissue and upper and lower epidermis were measured, and the variation characteristics of leaf structure after doubling were analyzed. Compared with diploid plants, the density of epidermal cells and stomatal apparatus in leaves of tetraploid plants changed significantly, and the density per unit field of vision significantly reduced due to the synchronous larging of epidermal cells and stomatal apparatus;the thickness of upper and lower epidermis, palisade tissue and sponge tissue in leaves of tetraploid plants were significantly larger than those of diploid plants.
