UBE2C和UBE2T与胰腺癌患者预后及免疫细胞浸润的关系 被引量:3

Relationships between UBE2C and UBE2T expression and prognosis and immune cells infiltration in pancreatic cancer

摘要 目的探讨泛素结合酶E2C(UBE2C)和E2T(UBE2T)与胰腺癌患者预后及免疫细胞浸润的关系。方法检索Oncomine、GEPIA2.0、EMBL-EBI数据库分析两基因在胰腺癌中的表达情况,利用GEPIA、Kaplan-Meierplotter数据库探索两基因的表达与胰腺癌患者预后的相关性。利用Oncomine及cBioPortal数据库筛选共表达基因,使用FunRich软件进行富集分析。利用String数据库和Cytoscape软件综合分析与两基因相互作用的蛋白及调控网络。使用Cytoscape筛选核心(Hub)基因,并利用GEPIA进行预后分析。使用TIMER数据库分析两基因的表达与免疫细胞浸润的相关性。结果UBE2C和UBE2T在胰腺癌中表达上调,高表达两基因的患者总生存期和无病生存期显著缩短(P均<0.05)。两基因的表达与患者的分期、分级、巨噬细胞浸润等有关(P均<0.05)。在胰腺癌中UBE2C与UBE2T的表达显著相关(P<0.05),与两基因共表达的基因主要富集于丝氨酸/苏氨酸蛋白激酶激活功能;主要参与细胞周期、纺锤体组装过程;主要位于染色体结构中;在细胞周期、Aurora-B信号通路中富集。筛选的10个Hub基因为PBK、CEP55、BIRC5、MELK、CDK1、NUSAP1、TPX2、BUB1、TOP2A和BUB1B,与胰腺癌患者预后相关(P<0.05)。两基因的表达与巨噬细胞浸润呈负相关关系(P<0.05)。结论UBE2C和UBE2T与胰腺癌患者的预后及多种免疫细胞如T细胞、巨噬细胞的浸润有关,可能通过调控细胞周期、DNA复制过程进而调控胰腺癌的进程。 Objective To analyze the ubiquitin-conjugating enzyme E2C(UBE2C)and ubiquitin-conjugating enzyme E2T(UBE2T)expression and prognosis and immune cells infiltration in pancreatic cancer patients.Methods Oncomine,GEPIA2.0,and EMBL-EBI databases were used to analyze the expression of UBE2C and UBE2T in pancreatic cancer.GEPIA and Kaplan-Meier plotter databases were used to analyze the correlations between gene expression and the prognosis of pancreatic cancer patients.The co-expressed genes were screened out based on Oncomine and cBioPortal databases.FunRich software was employed to explore the functional classification of co-expressed genes.String database and Cytoscape software were used to construct a protein-protein network.The Hub genes was screened out by using Cytoscape and the further analysis of prognosis was also performed by GEPIA database.We used the TIMER database to analyze the correlation between the expression of the two genes and the infiltration of immune cells.Results The expression of UBE2C and UBE2T in pancreatic cancer was up-regulated.Higher UBE2T and UBE2C expression tended to present shorter overall survival or disease-free survival of pancreatic cancer patients.The expression of UBE2C and UBE2T was related to the pathological stage,grade and macrophages infiltration(all P<0.05).UBE2C and UBE2T were highly correlated in the pancreatic cancer(P<0.05).A total of 59 co-expressed genes were mainly enriched in activated serine/threonine protein kinase molecular biological functions,were mainly involved in cell cycle and spindle assembly process,were mainly located in chromosomes,and were mainly enriched in cell cycle and Aurora-B signaling pathways(all P<0.05).Ten Hub genes PBK,CEP55,BIRC5,MELK,CDK1,NUSAP1,TPX2,BUB1,TOP2A,and BUB1B were screened out,which were related to the prognosis of pancreatic cancer patients(all P<0.05).The expression of UBE2C and UBE2T was negatively correlated with infiltration of macrophages(P<0.05).Conclusions The expression of UBE2C and UBE2T in pancreatic cancer is related to the prognosis of pancreatic cancer patients,and infiltration of T cells and macrophages in pancreatic cancer.The two genes may regulate the progression of pancreatic cancer by regulating cell cycle and DNA replication.
