摘要
目的:检测干眼模型兔泪腺组织中的p38MAPK信号通路中相关因子表达情况,初步探讨针刺对干眼的抗炎作用机制。方法:将48只健康纯种新西兰兔随机分为空白组、模型组、假针刺组、针刺组、西药组、针药组,共6组,每组8只。于不同时间段观察各组角膜荧光素钠染色评分(CFS)、泪膜破裂时间(BUT)及泪液分泌试验(SⅠT)情况,用ELISA法检测各组泪腺中乙酰胆碱(ACh)及受体α7烟碱型乙酰胆碱受体(α7nAChR)含量的变化,Western blot法检测泪腺中p38MAPK及p-p38MAPK的蛋白表达水平。结果:CFS、BUT、SⅠT等实验结果说明氢溴酸东莨菪碱造模21d有效,针刺、西药和针药结合治疗可减轻干眼模型兔角膜染色评分(P<0.05),延长BUT(P<0.05),增加泪液分泌量(P<0.05)。与空白组比较,模型组和假针刺组兔泪腺中ACh和α7nAChR含量均显著降低(P<0.05),p38MAPK及p-p38MAPK蛋白的表达量显著升高(P<0.05)。与模型组比较,针刺组、西药组和针药组兔泪腺中ACh和α7nAChR含量均显著上升(P<0.05),p38MAPK及p-p38MAPK蛋白的表达量显著降低(P<0.05)。结论:针刺对干眼的抗炎作用机制可能与胆碱能抗炎通路中的p38MAPK信号通路密切相关。
Objective: To investigate the effects of acupuncture on the related factors in the signaling pathway of p38 mitogen-activated protein kinase(MAPK) in the dry-eye model rabbits and to explore the anti-inflammatory mechanism of acupuncture on dry eye. Methods: Totally 48 healthy New Zealand white rabbits were randomly divided into 6 groups: blank group, model group, sham acupuncture group, acupuncture group, western medicine group, and acupuncture plus western medicine group, with 8 rabbits in each group. Six groups were examined for corneal fluorescin staining(CFS), break-up time of tear film(BUT) and Schirmer I test(SIT) results at different time points. ELISA was used to detect the contents of acetylcholine(ACh)and its receptor(α7 nAChR), and Western blot was used to detect the protein expression levels of p38 MAPK and p-p38 MAPK in the lacrimal glands of dry eye model rabbits. Results: The results of CFS, BUT and SIT indicated that scopolamine hydrobromide was effective for 21 days. Acupuncture treatment, western medicine treatment and acupuncture combined with western medicine treatment could reduce corneal fluorescin staining(P<0.05), prolong the BUT(P<0.05) and increase the amount of tear secretion of dry eye model rabbits(P<0.05). Compared with the blank group, the contents of ACh and α7 nAChR in the lacrimal glands of the model group and the sham acupuncture group were significantly reduced(P<0.05), and the expressions of p38 MAPK and p-p38 MAPK proteins in those groups were significantly increased(P<0.05). Compared with the model group, the contents of ACh and α7 nAChR in the lacrimal glands of the dry eye model rabbits were significantly increased in the acupuncture group, western medicine group and acupuncture plus western medicine group(P<0.05), and the expressions of p38 MAPK and p-p38 MAPK proteins in those groups were significantly reduced(P<0.05). Conclusion: The anti-inflammatory mechanism of acupuncture on dry eye may be closely related to the p38 MAPK signaling pathway.
