砂海星总蛋白提取及其酶解工艺条件优化 被引量:1

Extraction of Total Protein from Luidia Quinaria and Optimization of Enzymatic Hydrolysis Conditions

摘要 以砂海星为原料,采用裂解液水提取砂海星总蛋白,选用木瓜蛋白酶、胃蛋白酶、中性蛋白酶和碱性蛋白酶对海星总蛋白进行酶解获得最佳蛋白酶,再采用单因素实验和正交实验,探究最佳酶解时间、酶解温度、pH值和加酶量4个因素对砂海星蛋白水解度的影响。结果表明:最佳胃蛋白酶在pH为4,加酶量为2000 U/g、温度为37℃和时间为5 h时,砂海星总蛋白的最大水解度为86.5%,并且具有显著的清除自由基活性。 Using starfish(Luidia Quinaria von Martens)as raw material,the protein was extracted by lysate water,papain,pepsin,neutral protease and alkaline protease were used to hydrolyze the total protein of starfish.Single factor experiment and orthogonal experiment were used to explore the hydrolysis time,hydrolysis temperature,hydrolysis time and hydrolysis time.The effects of pH value and enzyme dosage on the degree of hydrolysis of starfish protein were studied.The results showed that the best pepsin was obtained.When pH was 4,the amount of enzyme was 2000 U/g,temperature was 37℃and time was 5 h,the maximum degree of hydrolysis of total protein of starfish was 86.5%,and it had significant free radical scavenging activity.
