摘要
[目的]探索桂郁金种质资源姜黄素类化学成分分析方法。[方法]:HPLC法被用以检测桂郁金姜黄素类成分,选用Agilent SB-C18色谱柱(250mm×4.6mm, 5μm),流动相为乙腈-冰醋酸水溶液,采用梯度洗脱,以420 nm作为检测波长,以30℃作为柱温;流速1 mL·min-1。[结果]桂郁金种质资源姜黄素类化学成分在测定范围内线性关系良好;符合要求的试验包括精密度、稳定性、重复性、加样回收试验。桂郁金姜黄素类化学成分的种类和含量由于种质不同而存在明显差异。[结论]该方法操作简单,适用于桂郁金种资源姜黄素类化学成分分析,其中桂郁金种质B71、C16在姜黄素类成分含量方面表现突出,为桂郁金优良种质。
To explore method for the analyzing curcumin in germplasm of Curcuma kwangsiensis,the HPLC method was used to detect the curcumin components in Curcuma kwangsiensis, Agilent SB-C18 chromatographic column(250 mm×4.6 mm, 5μm) was used, the mobile phase was taken as acetonitrile-glacial acetic acid solution, gradient elution was aplied, detect wave length was 420 nm, the column temperature was 30 30℃,the velocity was 1 ml/min. The results showed that the curcumin chemical constituents of germplasm of Curcuma kwangsiensis had good linear relationship in the determination range. The required tests include precision, stability, repeatability and recovery test. The types and contents of curcumin in Curcuma kwangsiensis were obviously different because of different germplasms. The method is simple to operate and suitable for the analyzing curcumin chemical constituents in the germplasm of Curcuma kwangsiensis.
