骨髓间充质干细胞对多发性骨髓瘤患者树突状细胞的影响

Effects of Bone Marrow-derived Mesenchymal Stem Cells on Dendritic Cells in Patients with Multiple Myeloma

摘要 【目的】探讨多发性骨髓瘤(MM)患者骨髓来源的间克质干细胞(MSC)对树突状细胞(DC)分化、成熟、细胞因子分泌以及DC辅助T细胞杀伤作用的影响。【方法】从MM患者和健康者骨髓获取MSC,与DC、T细胞及骨髓瘤细胞系U266细胞混合培养。采用流式细胞术检测DC免疫表型.AnnexinV-FITC/PI双标检测细胞凋亡情况。【结果】健康者MSC细胞培养液上清中转化生长因子β1为(60.52±12.83)pg/ml.明显低于MM患者的(302.51±70.52)pg/mL,且差异有显著性(P<0.05);MSC与未成熟/成熟DC共培养时。其CDla、CD11C、CD80、CD83、CD86、HLA-DR的表达明显抑制。同时减少白细胞介素-12分泌(P<0.05);T细胞+DC+U266混合培养中U266细胞凋亡率为(72.24±8.63)%,T细胞+DC+U266+MM患者MSC混合培养中U266细胞凋亡率为(45.56±9.22)%,T细胞+DC+U266+健康者MSC混合培养中U266细胞凋亡率为(25.53±7.31)%,三组相比较差异均有显著性(P<0.05)。【结论】MM患者MSC能明显抑制DC分化、成熟和细胞因子分泌,同时降低DC辅助T细胞杀伤作用。 【Objective】To investigate efeets of mutiple myeloma(MM)patients with bone marrow derived mesenchymal stem cell(MSC)on differentiation.maturation and cytokinese cretion of dendritic cell(DC)and its effeet on DC helper T cell skilling myeloma cells.【Methods】MSC cells from MM patients and healthy volunteers were mixed and cultured with DC,T cells and myeloma cell line U266 cells.DC immuno phenotype was detected by flow cytometry.and cell apoplosis were measured by Annexin V-FITC/PI double labeling.【Results】Culure supernatant TGFβ1 level of healthy persons,MSC cells was(60.52±2.83)pg/ml.which was significantly lower than that in MM patients(302.51±70.52)pg/ml;Tthe difference was statisti-cal significance(P<0.05).Co-culture of MSC and immature/mature DC cells showed that CD1a.CD11C.CD80,CD83.CD86,IL-12 secretion and HLA DR expression were significantly suppessed(P<0.05).When T cells+DC+U266 were cultured together.the apoptosis rate in U266 cells was(72.24±8.63)%;While co-culturing T cell+DC+U266+MSC from MM patients.the apoptosis rate in U266 cells was(45.56±9.22)%;When mixing T cell+DC,U266 cells with MSC from healthy persons.the apoptosis rate in U266 cells was(25.53±7.31)%.All differences were statistically significant(P<0.05).【Conclusion】MSC from MM patients can significantly inbibit DC differentiation.maturation and cytokine secretion.and inhibit the killing efect of DC helper T cells.
