春兰种质资源的EST-SSR遗传多样性分析及指纹图谱构建 被引量:5

Genetic Diversity Analysis and Construction of Fingerprints of Cymbidium goeringii Germplasms Based on EST-SSR Markers

摘要 为研究春兰种质的遗传多样性,本研究利用筛选出的19对具有多态性且条带清晰的引物,对48份春兰种质资源进行遗传多样性分析和指纹图谱构建。结果表明,19对多态性引物共检测到77个等位基因,平均每对引物检测到4.0526个等位基因;观测杂合度(Ho)平均为0.4375,期望杂合度(He)平均为0.5470;Shannon’s多样性信息指数(I)介于0.6579~1.4059之间,平均为0.9762;多态信息含量(PIC)变化范围为0.3281~0.6404,平均为0.4815;基因流(Nm)的平均值为12.6246。对3个参试群体进行遗传多样性分析,色花群体(SH)的遗传多样性最高;色花群体(SH)与素心花群体(SX)亲缘关系较近,与蝶花群体(DH)之间亲缘关系较远。48份春兰品种遗传相似系数变幅为0.71~1.00,在遗传相似度为0.745时,48个种质聚成5类。仅用5对核心引物能够区分参试种质,并为每份种质构建了指纹图谱。本研究结果可为今后春兰新品种选育及品种鉴定提供重要依据。 In order to study the geneti c diversity of Cymbidium goeringii germplasm,the genetic diversity and fingerprint map of 48 Cymbidium goeringii germplasm resources were studied using 19 pairs of selected polymorphic EST-SSR primers with polymorphism and clear bands.The results showed that a total of 77 alleles were detected,with an average of 4.052 6 alleles per locus.The mean values of observed heterozygosity( Ho) and expected heterozygosity(He) were 0.437 5 and 0.547 0,respectively.Shannon’s diversity information index(I) ranged from0.657 9 to 1.405 9,with an average value of 0.976 2.The polymorphic information content(PIC) ranged from0.328 1 to 0.640 4,with an average value of 0.481 5.The mean value of gene flow(Nm) was 12.624 6.Genetic diversity analysis of the three populations showed that the population of colored flower(SH) had the highest genetic diversity.The population of colored flower(SH) was close to the population of concolor petal(SX),and far away from the population of butterfly petal(DH).Genetic similarity coefficient of 48 Cymbidium goeringii varieties ranged from 0.71 to 1.00.48 germplasms were grouped into five groups at the genetic similarity coefficient of0.745.5 pairs of primers were able to distinguish the test germplasms,and each variety had its unique fingerprint.These results can provide important basis for variety breeding and Cymbidium goeringii identification in the future.
