摘要
目的研究益肾强身丸和防衰益寿丸对雄性小鼠生殖功能的改善作用。方法将48只2月龄C57BL/6J小鼠随机分为对照组,益肾强身丸+防衰益寿丸低剂量组(YS 0.25 g·kg^(-1)+FS 0.25 g·kg^(-1)组)、高剂量组(YS 0.5 g·kg^(-1)+FS 0.5 g·kg^(-1)组),五子衍宗丸1 g·kg^(-1)组(简称阳性药组)。高、低剂量组每日上午灌胃对应剂量的益肾强身丸溶液,下午灌胃对应剂量的防衰益寿丸溶液;对照组及阳性药组每日上午灌胃相应的饮用水和五子衍宗丸,连续30 d。记录小鼠7、14、21、28 d跨骑次数、相关脏器指数、精子数目和活性、血清睾酮、海绵体组织NO和cGMP水平。结果与对照组比较,低剂量组可提高小鼠性行为次数(P<0.01),低、高剂量组均可增加精子数目、提高精子活率及活力且降低精子畸形率(P<0.01,P<0.001);高剂量组可提高海绵体cGMP水平(P<0.001);低、高剂量组均对性器官指数没有影响。结论益肾强身丸和防衰益寿丸可改善雄性小鼠的生殖功能。
Objective To determine the effect of Yishen Qiangshen pills and Fangshuai Yishou pills on the reproductive function in male mice.Methods Totally 48 two-month-old C57BL/6J mice were randomly divided into 4 groups,including a control group,a YS 0.25 g·kg^(-1)+FS 0.25 g·kg^(-1) group(the low dose group),a YS 0.5 g·kg^(-1)+FS 0.5 g·kg^(-1) group(the high dose group)and a WZYZ 1 g·kg^(-1) group(positive group).Both the low and the high dose groups were given Yishen Qiangshen pills intragastrically in the morning and Fangshuai Yishou pills in the afternoon.The control group and the positive group were given constant volume of water or Wuzi Yanzong pills intragastrically in the morning for 30 days consecutively.The total number of intercourses within 20 min was recorded on day 7,14,21 and 28 after the treatment.The related organ indexes were also measured,and the number and viabilities of sperms in each group were detected.After the intervention,the concentration of serum testosterone and the level of NO and cGMP in corpus cavernosum tissue were measured.Results As compared with the control group,the total number of intercourses was increased within 20 min(P<0.01)in the low dose group.The number and viabilities of sperms were significantly increased and the rate of sperm malformation decreased(P<0.01,P<0.001)in the low and the high dose groups.The level of cGMP in the corpus cavernosum tissue was increased in the high dose group(P<0.001).In addition,both the low and high dose had no effect on the sexual organ indexes.Conclusion Yishen Qiangshen pills and Fangshuai Yishou pills could improve the reproductive function in male mice.
