柔肝化纤颗粒动员骨髓间充质干细胞归巢治疗肝纤维化大鼠的机制研究 被引量:6

摘要 目的探讨柔肝化纤颗粒对大鼠骨髓间充质干细胞(BMSCs)归巢于肝脏的影响及治疗肝纤维化效果。方法选取SD雄性大鼠52只,分为空白对照组(10只)、模型组(42只),建模成功后将肝纤维化大鼠模型随机分为模型组、BMSCs组、柔肝化纤颗粒组、柔肝化纤颗粒+BMSCs组。干预4周后,分别采用qPCR、Western Blot检测各组肝组织AFP、ALB、CK18基因表达与SDF-1、CXCR4、HGF、EGF基因及蛋白表达、HE染色、Masson染色检测肝组织纤维化程度、PAS法检测肝脏储存糖原功能及冷冻切片检测携带绿色荧光蛋白的BMSCs向肝组织归巢情况。结果动物实验方面,与模型组相比,使用柔肝化纤颗粒联合BMSCs移植组肝脏组织AFP、ALB、CK18表达上调,HGF、EGF表达上调,肝脏内携带绿色荧光蛋白的BMSC数量明显增加,肝纤维化程度下降。结论柔肝化纤颗粒能动员BMSCs归巢于纤维化肝组织,并能够修复受损肝组织,其机制可能与SDF-1/CXCR4轴有关。 Objective To investigate the effect of Rougan Huaxian Granules(柔肝化纤颗粒)on the homing of rat bone mar-row mesenchymal stem cells(BMSCs)in liver and the therapeutic effect of liver fibrosis.s Methods A total of 52 SD male rats were selected and divided into blank control group(10 rats)and model group(42 rats).After successful modeling,rat models of liver fibrosis were randomly divided into model group,BMSCs group,Rougan Huaxian Granules group,Rougan Huaxian Granules+BMSCs group.After 4 weeks intervention,The expressions of AFP,ALB,CK18 as well as gene and protein expressions of SDF-1,CXCR4,HGF and EGF were detected by qPCR and Western Blot.The degree of liver fibrosis was detected by HE staining and Masson staining.The glycogen storage function of liver was detected by PAS method.The homing of BMSCs carrying green fluorescent protein to liver tissue was detected by frozen section.s Results In animal experiments,compared with those of the mod-el group,the expressions of AFP,ALB and CK18,HGF and EGF in the liver tissues of the BMSCs transplantation group were up-regulated,the number of BMSC carrying green fluorescent protein in the liver was significantly increased,and the degree of liver fibrosis was decreased.n Conclusion Rougan Huaxian Granules can mobilize BMSCs to nest in fibrosis liver tissue and repair damaged liver tissue,and the mechanism may be related to SDF-1/CXCR4 axis.
