牛磺酸缓解H_(2)O_(2)诱导的鸡胚肝细胞氧化损伤 被引量:1

摘要 为探索牛磺酸对H_(2)O_(2)诱导的鸡胚肝细胞氧化损伤的缓解作用,文章用不同浓度的H_(2)O_(2)作用于鸡胚原代肝细胞,以细胞相对存活率、胞内丙二醛(MDA)产生量、超氧化物歧化酶(SOD)活性和过氧化氢酶(CAT)活性等指标作为判断依据,建立稳定的鸡胚原代肝细胞氧化损伤模型;通过形态观察、牛磺酸安全浓度检测和细胞活力检测等方法研究牛磺酸对鸡胚氧化损伤的缓解作用。结果表明,以H_(2)O_(2)为刺激源处理鸡胚原代肝细胞4 h,H_(2)O_(2)浓度为8 mmol/L时,细胞存活率接近60%,MDA产生量、SOD、CAT活性相对于空白对照明显增加。安全浓度检测结果表明,牛磺酸对鸡胚肝细胞的安全浓度为6.25、12.50 mmol/L;细胞活力检测结果表明,在牛磺酸浓度为12.50 mmol/L时细胞的存活率接近80%。实验结果表明,牛磺酸可以显著抑制H_(2)O_(2)对鸡胚肝细胞的氧化损伤,为深入研究鸡胚抗氧化机理奠定一定的基础。 In order to explore the alleviative effect of taurine on oxidative damage of chicken embryo hepatocytes induced by hydrogen peroxide(H_(2)O_(2)),chicken embryo primary hepatocytes were treated with different concentrations of H_(2)O_(2).A stable oxidative damage model of chicken embryo primary hepatocytes was established based on the indexes such as cell relative survival rate,intracellular malondialdehyde(MDA)content,superoxide dismutase(SOD)and catalase(CAT)activities.The alleviative effect of taurine on oxidative damage of chicken embryo was studied by morphological observation,taurine safety concentration detection and cell viability detection.When the concentration of H_(2)O_(2)was 8 mmol/L,the cell survival rate was close to 60%after a 4 h treatment,and the MDA content and the activities of SOD and CAT were significantly increased compared with those of the blank control group.The safe concentration of taurine to chicken embryo hepatocytes was 6.25 mmol/L and 12.50 mmol/L.The cell viability test showed that the cell survival rate was close to 80%when taurine concentration was 12.50 mmol/L.The experimental results showed that taurine could significantly inhibit the oxidative damage of chicken embryo hepatocytes caused by H_(2)O_(2),which laid a foundation for further study on the antioxidant mechanism of chicken embryo.
