miR-20a在肝癌中的研究进展

摘要 国内外研究显示miR-20a在多种肿瘤中异常表达,其中在肝癌方面研究较为广泛并且发现miR-20a在肝癌组织中过低表达,这种表现在肝癌的产生、进展、转移过程充当重要角色。miR-20a与肝癌患者的预后有关,miR-20a越低预后越差。过低的miR-20a促使抗凋亡成员Mcl-1显著升高进而抑制癌细胞凋亡。与抑癌基因PTEN有高度同源性的假基因PTENP1可隔离miR-20a从而抑制PI3K/AKT信号通路,诱导促死亡自噬,抑制肝癌细胞在体内的生长,并伴有细胞凋亡、自噬和抑制血管形成及成熟。但是高表达的miR-20a可借助激活PTEN/PI3K/AKT途径致使肝肿瘤产生放射抗性。高表达的miR-20a通过靶向CCND1抑制着肝癌细胞系HepG2细胞凋亡、迁移和侵袭。miR-20a可以抑制CCND1的过度表达,从而达到治疗肝癌的目的。因而miR-20a可望作为诊断肝癌的标记物及治疗肝癌的靶点。 Studies at home and abroad show that miR-20a expresses abnormally in a variety of tumors.For instance,the low expression of miR-20a in liver cancer tissue has been widely discussed in literature.Especially,miR-20a plays an important role in the production,progression and metastasis of liver cancer,and the lower miR-20a is associated with the worse prognosis of liver cancer patients.Low miR-20a prompts a significant increase of the anti-apoptotic Mcl-1,which subsequently inhibits the apoptosis of cancer cells.PTENP1,a pseudogene with high homology to the tumor suppressor gene PTEN,can isolate miR-20a,thereby inhibiting the PI3 K/AKT signaling pathway and inducing pro-death autophagy.The process can inhibit the growth of liver cancer cells in vivo,accompanied by apoptosis,autophagy and inhibition of angiogenesis and maturation.However,high expression of miR-20a can induce the radio-resistance of hepatoma cells by activating PTEN/PI3 K/AKT pathway.High expression of miR-20a inhibits apoptosis,migration and invasion of HepG2 in hepatoma cell line by targeting CCND1.miR-20a can also inhibit the overexpression of CCND1 and thus achieve the purpose of treating liver cancer.Therefore,miR-20a is expected to be a marker for the diagnosis of liver cancer and a target in treatment.
