摘要
目的:探究桂枝茯苓胶囊对子宫肌瘤大鼠细胞增殖、迁移和侵袭、MAPK信号通路和T淋巴细胞水平的影响。方法:60只大鼠随机分为对照组、子宫肌瘤组和子宫肌瘤+GZFLc组(n=20)。通过注射苯甲酸雌二醇和黄体酮构建子宫肌瘤模型。桂枝茯苓胶囊灌胃干预,剂量为1 000 mg·kg^(-1)。检测各组小鼠激素水平、子宫系数、雌二醇(E2)、孕酮(P)、促卵泡激素(FSH)、细胞间黏附分子1(ICAM1)、附睾蛋白4(HE4)、Ki67、基质金属蛋白酶2(MMP2)、ERK1/2、p38 MAPK蛋白表达量。通过流式细胞术检测CD3^(+)比例和CD4^(+)/CD8^(+)水平。结果:3组大鼠上述指标比较差异有统计学意义(P<0.05)。子宫肌瘤组大鼠血清中E2[(782.45±65.38) pg·mL^(-1)]、P[(57.11±5.84) ng·mL^(-1)]和FSH[(6.24±0.61) IU·L^(-1)]、子宫系数(15.36±1.92)、ICAM1(1.82±0.15)、HE4(1.19±0.12)、Ki67(0.74±0.07)、MMP2(0.62±0.06)、ERK1/2(1.28±0.11)、p38 MAPK(1.10±0.10)水平显著高于对照组,CD3^(+)[(34.58±2.35)%]和CD4^(+)/CD8^(+)(1.17±0.22)水平显著低于对照组(P<0.05)。子宫肌瘤+GZFLc组的E2[(437.62±41.49) pg·mL^(-1)]、P[(46.52±4.57) ng·mL^(-1)]、FSH[(4.85±0.46) IU·L^(-1)]、子宫系数(8.25±1.04)、ICAM1(0.76±0.07)、HE4(0.71±0.07)、Ki67(0.32±0.03)、MMP2(0.27±0.03)、ERK1/2(0.52±0.05)和p38 MAPK(0.56±0.05)水平显著低于子宫肌瘤组,CD3^(+)(42.44±3.76)和CD4^(+)/CD8^(+)(1.85±0.31)水平显著高于子宫肌瘤组(P<0.05)。结论:桂枝茯苓胶囊可抑制子宫肌瘤的增殖迁移和侵袭,抑制MAPK通路,提高CD3^(+)、CD4^(+)/CD8^(+)的水平。
OBJECTIVE To explore the effects of Guizhi Fuling Capsule on cell proliferation, migration and invasion, MAPK signaling pathway and T lymphocyte levels in rats with uterine fibroids.METHODS Sixty rats were randomly divided into three groups of control, uterine fibroids and uterine fibroids+GZFLc(n=20).The model of uterine fibroids was constructed by injecting estradiol benzoate and progesterone.Guizhi Fuling Capsules were administered intragastrically at a dose of 1 000 mg·kg^(-1).Hormone levels, uterine coefficient, estradiol(E2),progesterone(P),follicle stimulating hormone(FSH),intercellular adhesion molecule 1(ICAM1),epididymal protein 4(HE4),Ki67,matrix metalloproteinase 2(MMP2),ERK1/2 and p38 MAPK proteins were measured.And CD3^(+) ratio and CD4^(+)/CD8^(+) level were detected by flow cytometry.RESULTS The above parameters of three groups were significantly different(P<0.05).Serum E2[(782.45±65.38) pg·mL^(-1)],P[(57.11±5.84) ng·mL^(-1)],FSH[(6.24±0.61) IU·L^(-1)],uterine coefficient(15.36±1.92),ICAM1(1.82±0.15),HE4(1.19±0.12),Ki67(0.74±0.07),MMP2(0.62±0.06),ERK1/2(1.28±0.11)and p38 MAPK(1.10±0.10)were significantly higher in uterine fibroids group than those in control group.CD3^(+)[(34.58±2.35)%]and CD4^(+)/CD8^(+)(1.17±0.22)were significantly lower than those in control group(P<0.05).E2[(437.62±41.49) pg·mL^(-1)],P[(46.52±4.57) ng·mL^(-1)],FSH[(4.85±0.46) IU·L^(-1)],uterine coefficient(8.25±1.04),ICAM1(0.76±0.07),HE4(0.71±0.07),Ki67(0.32±0.03),MMP2(0.27±0.03),ERK1/2(0.52±0.05)and p38 MAPK(0.56±0.05)were significantly lower in uterine fibroids+GZFLc group than those in uterine fibroids group, CD3^(+)(42.44±3.76)and CD4^(+)/CD8^(+)(1.85±0.31)levels were significantly higher than those in uterine fibroids group(P<0.05).CONCLUSION Guizhi Fuling Capsule can suppress the proliferation, migration and invasion of uterine fibroids, inhibit the MAPK pathway and boost the levels of CD3^(+)and CD4^(+)/CD8^(+).
