南海近海浮游细菌群落结构和多样性研究 被引量:4

Diversity of pelagic bacterial community structure in the South China Sea

摘要 为了解南海东北部、琼东和北部湾3处水域内浮游细菌的群落结构和多样性特征,利用Illumina Miseq高通量测序技术对40个站位64个细菌样品的16S rRNA基因V3-V4高变区进行测序分析。在97%序列相似性水平条件下,调查区域内总共鉴定2891个操作分类单元(operational taxonomic unit,OTU),单样品OTU个数变化范围为249~1250。鉴定OTU隶属于37个细菌门、101个纲、243个目、404个科、753个属(包含未分类部分)。变形菌门(Proteobacteria)、蓝细菌门(Cyanobacteria)、放线菌门(Actinobacteria)和拟杆菌门(Bacteroidota)是4个最具数量优势的细菌门,分别占研究采集样品序列总数的39.24%、29.67%、10.97%和10.80%;蓝细菌纲(Cyanobacteria)、α-变形菌纲(Alphaproteobacteria)、γ-变形菌纲(Gammarproteobacteria)、拟杆菌纲(Bacteroidia)和酸微菌纲(Acidimicrobiia)是样品中5个具数量优势的细菌纲,序列数占比分别为29.67%、22.76%、16.47%、10.71%和8.52%;聚球藻属(Synechococcus)、原绿球藻属(Prochlorococcus)、Candidatus_Actinomarina、蓝细菌属(Cyanobium)为数量占优的细菌属。Wilcoxon秩和检验表明,研究区域表层水体浮游细菌群落结构没有显著的空间差异,南海东北部水域群落多样性通常较高,显示珠江河口淡水注入以及上升流等物理事件在区域内营造了较为异质化的生境。浮游细菌群落呈现较为显著的层化现象,底层群落具有更高的多样性和特异性生物类群。 To understand the pelagic bacterial community structure and distribution characteristics in the northeast of South China Sea,east Hainan Island and Beibu Gulf,the V3-V4 hypervariable regions of bacterial 16S rRNA gene in 64 samples from 40 stations were sequenced by Illumina Miseq high-throughput sequencing technology.A total of 2891 operational taxonomic units were identified at 97%sequence similarity level(the number of OTUs in each sample ranged from 249 to 1250).The OTUs were identified to belong to 37 phylum,101 classes,243 orders,404 families and 753 genera.Proteobacteria,Cyanobacteria,Actinobacteria and Bacteroidota were the four most numerically dominated bacteria in seawater samples,which accounted for 39.20%,29.33%,11.00%and 10.62%of total sequences,respectively.Cyanobacteria(29.67%),α-Proteobacteria(22.76%),γ-Proteobacteria(16.47%),Bacteroidia(10.71%)and Acidimicrobiia(8.52%)were the five most numerically dominated bacterial classes.Synechococcus,Prochlorococcus,Candidatus_Actinomarina and Cyanobium were the numerically dominated bacterial genera.Wilcoxon rank-sum test showed that no significant spatial difference was found in the pelagic bacterial community structure among the studied areas.The diversity indices of pelagic bacterial community in the northeast South China Sea were much higher than those of other studied areas,which was considered to be closely related to the heterogeneous habitats resulted from the riverine input from the Pearl River Estuary and hydrodynamic factors such as upwelling events.Significant discrepancies were observed in bacterial communities between surface and bottom waters.Higher diversity and richness index were commonly present in surface layers in the studied area.
