摘要
目的:观察青紫颗粒对麦胶蛋白致小鼠肾损伤的保护作用。方法:84只SPF级KM小鼠随机分为7组:正常对照组、模型组、青紫颗粒低(4.11 g·kg^(-1))、中(8.22 g·kg^(-1))、高剂量组(16.44 g·kg^(-1))、地塞米松组(0.63 mg·kg^(-1))、芦丁片组(12.54 mg·kg^(-1)),每组12只。除正常对照组外,其余各组采用口服麦胶蛋白抗原法建立肾损伤小鼠模型。从第15周开始,各给药组灌胃给与相应药物,1次/d,连续3周,正常对照组和模型组灌胃相同体积蒸馏水。采用ELISA法检测血清尿素氮(BUN)、肌酐(Cr)、晚期氧化蛋白产物(AOPPs)、循环免疫复合物(CIC)水平;免疫荧光法检测肾脏单纯免疫球蛋白A(IgA)沉积,PAS染色法观察肾组织IgA_(1)异常糖基化情况,免疫荧光法检测小鼠肾脏补体C3、C4及转化生长因子β_(1)(TGF-β_(1))的表达,HE染色法观察肾组织病理学变化。结果:与正常对照组相比,模型组血清BUN、Cr、AOPPs、CIC水平显著升高(P<0.05或P<0.01),肾脏IgA、补体C4及TGF-β_(1)表达显著增加(P<0.05或P<0.01),肾组织出现IgA_(1)异常糖基化,病理损伤明显。青紫颗粒可降低麦胶蛋白所致肾损伤小鼠血清BUN、Cr、AOPPs、CIC水平(P<0.05或P<0.01),下调肾脏IgA、补体C4及TGF-β_(1)表达(P<0.05或P<0.01),减轻IgA_(1)异常糖基化及肾脏病理损伤。结论:青紫颗粒对麦胶蛋白所致肾损伤小鼠具有保护作用,该作用可能与其降低AOPPs、CIC水平,下调IgA、补体C4及TGF-β_(1)表达,减轻IgA_(1)异常糖基化有关。
Objective:To observe the protective effect of Qingzi granules on gliadin-induced kidney injury in mice.Methods:Totally 84 SPF KM mice were randomly divided into 7 groups:normal control group,model group,low(4.11 g·kg^(-1)),medium(8.22 g·kg^(-1))and high-dose Qingzi granules groups(16.44 g·kg^(-1)),dexamethasone group(0.63 mg·kg^(-1))and rutin tablets group(12.54 mg·kg^(-1))with 12 rats in each group.Except the normal control group,the other groups used oral gliadin antigen method to establish kidney injury mouse models.From the 15 th week,each administration group was given the corresponding drug by gavage,once a day,for 3 consecutive weeks,and the normal control group and the model group were given the same volume of distilled water.ELISA method was used to detect serum blood urea nitrogen(BUN),creatinine(Cr),advanced oxidized protein products(AOPPs)and circulating immune complex(CIC)levels.Immunofluorescence method was used to detect kidney pure immunoglobulin A(IgA)deposition,PAS staining method was used to observe renal tissue IgA1 abnormal glycosylation,immunofluorescence method was used to detect mouse kidney complement C3,C4 and transforming growth factorβ1(TGF-β1),and HE staining method was used to observe the pathological changes of kidney tissue.Results:Compared with those of the normal control group,the serum levels of BUN,Cr,AOPPs and CIC in the model group were significantly increased(P<0.05 or P<0.01),and the expression levels of kidney IgA,complement C4 and TGF-β1 were significantly increased(P<0.05 or P<0.01);abnormal glycosylation of IgA1 occurred in the kidney,and the pathological damage of kidney tissue was obvious.Qingzi granules could reduce the serum BUN,Cr,AOPPs,and CIC levels(P<0.05 or P<0.01),reduce the expression levels of kidney IgA,complement C4 and TGF-β1(P<0.05 or P<0.01),and reduce the abnormal glycosylation of kidney IgA1 and alleviate the pathological damage of the kidney in HSP mice induced by gliadin.Conclusion:Qingzi granules can alleviate the renal damage of HSP mice caused by gliadin,and this effect is related to reducing the levels of AOPPs and CIC,down-regulating the expression levels of IgA,C4 and TGF-β1,and decreasing abnormal glycosylation of IgA1.
