摘要
目的:研究长链非编码RNA CASC2在肝癌中的表达及其临床意义,并探讨其作用机制。方法:采用RT-PCR检测42例肝癌患者肿瘤组织中CASC2的表达水平,分析CASC2表达对肝癌患者各临床病理参数和预后的影响。通过细胞转染建立CASC2过表达的肝癌细胞系,采用CCK-8法检测各组细胞的增殖活性,Transwell法检测细胞迁移和侵袭能力。双荧光素酶实验验证CASC2与miR-221的相互作用。并采用RT-PCR检测各组细胞和肝癌组织中miR-221的表达水平。在CASC2组细胞中过表达miR-221,观察细胞增殖、迁移和侵袭能力的变化。结果:CASC2在肝癌患者肿瘤组织中表达水平显著低于癌旁正常组织(P<0.05)。通过分析发现,CASC2表达水平与肝癌患者的TNM分期和血管侵犯具有显著相关性(均P<0.05),且CASC2高表达者具有更长的总生存周期。过表达CASC2后,肝癌细胞的增殖、迁移和侵袭能力显著下降(均P<0.05),CASC2可直接靶向作用miR-221并抑制其表达。逆转实验结果表明,过表达miR-221后Huh7细胞的增殖、迁移和侵袭能力均显著增强(均P<0.05)。结论:长链非编码RNA CASC2与肝癌患者的TNM分期、血管侵犯及预后具有相关性,其可通过调控miR-221表达抑制肝癌细胞的增殖、迁移和侵袭。
Objective:To investigate the expression of long non-coding RNA CASC2 in liver cancer and its clinical significance,and to explore its mechanism.Methods:The CASC2 expression in tumor tissues of 42 liver cancer patients were detected by RT-PCR.The influence of CASC2 on clinicopathological parameters and prognosis of patients with liver cancer were analyzed.The liver cancer cell line overexpressing CASC2 was established by cell transfection method.The CCK-8 method was used to detect cell proliferation.Transwell method was used to detect cell migration and invasion.The targeted binding of CASC2 to miR-221 was detected by dual luciferase method.The expression level of miR-221 in cells and liver cancer tissues were detected by RT-PCR.The cell proliferation,migration and invasion ability were detected after miR-221 overexpression in CASC2 group.Results:CASC2 expression in tumor tissues of liver cancer patients were significantly lower than that of normal tissues adjacent to the tumor(P<0.05).The CASC2 expression was significantly correlated with TNM staging and vascular invasion(all P<0.05).Patients with high CASC2 expression have a longer overall survival.The proliferation,migration and invasion ability of liver cancer cells were decreased significantly after overexpression of CASC2(all P<0.05).The dual luciferase assay suggested that CASC2 can directly target miR-221 and inhibit its expression.After overexpression of miR-221,the proliferation,migration and invasion ability of Huh7 cells were significantly enhanced(all P<0.05).Conclusion:The long non-coding RNA CASC2 is correlated to the TNM staging,vascular invasion and prognosis of patients with liver cancer,which could inhibit proliferation,migration and invasion of liver cancer cells by regulating the expression of miR-221.
