子痫前期患者胎盘组织miR-373、PDCD4表达与胎盘螺旋动脉重铸的关系 被引量:3

The relationship between the expression of miR-373 and PDCD4 in placental tissues of preeclampsia andplacental spiral artery remodeling

摘要 目的检测微小RNA-373(miR-373)、程序性细胞死亡因子4(PDCD4)在子痫前期患者胎盘组织中的表达水平,探讨miR-373、PDCD4表达与胎盘螺旋动脉重铸的关系。方法选择2019年4月至2020年11月在恩施州中心医院产科建卡并首次诊断为子痫前期的43例孕妇为研究对象(子痫前期组),另选正常妊娠孕妇45例作为对照组,采用实时荧光定量PCR(RT-qPCR)法检测胎盘组织miR-373、PDCD4mRNA表达水平,免疫印迹法检测胎盘组织中PDCD4蛋白表达;Pearson法分析胎盘组织miR-373、PDCD4表达水平与螺旋动脉管壁厚度及管腔面积的相关性。结果子痫前期组入院收缩压与舒张压显著高于对照组(P<0.001);两组年龄、体重指数及分娩孕周比较,差异无统计学意义(P>0.05)。与对照组比较,子痫前期组胎盘组织中miR-373表达水平、平均管腔面积显著降低,PDCD4mRNA及蛋白表达水平、平均管壁厚度显著升高,差异具有统计学意义(P<0.001)。子痫前期患者胎盘组织中miR-373与管壁厚度呈负相关(r=-0.411,P=0.006),与管腔面积呈正相关(r=0.547,P<0.001);PDCD4与管壁厚度呈正相关(r=0.439,P=0.003),与管腔面积呈负相关(r=-0.663,P<0.001)。结论子痫前期患者胎盘组织中miR-373低表达,PDCD4高表达,两者均与子痫前期胎盘螺旋动脉重铸障碍过程有关。 Objective To detect the expression levels of microRNA-373(miR-373) and programmed cell death 4(PDCD4) in placental tissues of preeclampsia, and explore the relationship between miR-373, PDCD4 expression and placental spiral artery remodeling. Methods 43 pregnant women who were diagnosed with preeclampsia in the Department of Obstetrics of Enshi Central Hospital from April 2019 to November 2020 were selected as the research objects(preeclampsia group), and 45 pregnant women with normal pregnancy were selected as the control group. Real-time fluorescent quantitative PCR(RT-qPCR) method was used to detect the expression levels of miR-373 and PDCD4 mRNA in placental tissues. Western Blotting method was used to detect PDCD4 protein expression in placenta tissue;Pearson method was used to analyze the correlation between the expression levels of miR-373 and PDCD4 in placental tissues and the thickness of spiral artery wall and lumen area. Results Systolic blood pressure and diastolic blood pressure in preeclampsia group were significantly higher than those in control group(P<0.001), and there were no significant differences in age, body mass index and gestational age between the two groups(P>0.05). Compared with the control group, the expression level of miR-373 and average lumen area in placental tissues of preeclampsia group were significantly decreased, while the expression level of PDCD4 mRNA and protein and average tube wall thickness were significantly increased(P<0.001). MiR-373 was negatively correlated with tube wall thickness(r=-0.411, P=0.006) and positively correlated with lumen area(r=0.547, P<0.001) in placental tissues of patients with preeclampsia. PDCD4 was positively correlated with tube wall thickness(r=0.439, P=0.003) and negatively correlated with lumen area(r=-0.663, P<0.001). Conclusions The low expression of miR-373 and high expression of PDCD4 in the placental tissues of preeclampsia patients are associated with the process of defective recasting of the placental spiral artery in preeclampsia.
