浊点萃取/超高效液相色谱-串联质谱法测定还亮草中五种生物碱

摘要 建立了浊点萃取技术结合超高效液相色谱-串联质谱法(UPLC-MS/MS),同时测定还亮草中5种生物碱含量的分析方法。采用单因素实验考察了浊点萃取中各影响因素对还亮草中5种生物碱萃取率的影响,确定最佳萃取条件为:4%的Trion X-114,NaCl的用量为0.4 g,溶液pH值为8,平衡温度55℃,平衡时间25 min。以乙腈-0.1%三乙胺为流动相进行UPLC-MS/MS分析,5种生物碱之间具有良好的分离度,且各成分在一定质量浓度范围内具有良好的线性关系。方法的精密度(RSD)、重复性及加标回收率的RSD均小于3.0%。该方法简单快捷、灵敏准确,可满足还亮草中生物碱检测的要求。 A method base on cloud point extraction/UPLC-MS/MS was developed for extraction and determination of five alkaloids in Delphinium anthriscifolium Hance.Experimental conditions affecting the cloud point extraction of five alkaloids were optimized.The optimum conditions were 4.0% Triton X-114,0.4 g NaCl,pH=8,55 ℃ of extraction temperature,and 25 min of extraction time.With acetonitrile-0.1% triethylamine as the mobile phase,five alkaloids were separated.The RSDs of recovery,precision,and reproducibility were all less than 3.0%.The results showed that cloud point extraction was suitable for the extraction of five alkaloids in Delphinium anthriscifolium Hance.
