摘要
为了探究脂肪间充质干细胞(AD-MSCs)对犬急性肾损伤(AKI)的治疗效果,将6只健康中华田园犬随机分为模型组和治疗组,每组3只,采用注射庆大霉素建立犬AKI模型,造模成功后,治疗组静脉注射AD-MSCs,对照组注射等量生理盐水,通过观察临床症状、检测尿常规及血清肌酐(SCr)、尿素氮(BUN)及肿瘤坏死因子-α(TNF-α)、中性粒细胞明胶酶相关脂质运载蛋白(NGAL)、肾损伤分子-1(KIM-1)来评估AD-MSCs的治疗效果,并在试验结束时取肾组织制备病理切片进行观察。结果显示,与造模前相比,庆大霉素可导致SCr和BUN大幅度升高,TNF-α、NGAL、KIM-1也持续升高,尿常规异常。经AD-MSCs治疗后,与模型组相比,治疗组SCr、BUN和TNF-α水平降低,NGAL和KIM-1恢复至正常水平;尿常规恢复正常;切片结果显示,治疗组的肾小管上皮细胞的损伤、坏死明显少于模型组。结果表明静脉注射犬AD-MSCs对犬AKI有一定治疗作用。
This study was undertaken to explore the therapeutic potential of adipose derived mesenchymal stem cells(AD-MSCs)for acute kidney injury(AKI)in canine.Six healthy Chinese garden dogs were randomly divided into model group and treatment group with 3 dogs each.Gentamicin injection was used to establish canine AKI model.Afterwards the treatment group was intravenously injected with AD-MSCs,while the control group was injected with the same amount of normal saline.The therapeutic effect of AD-MSCs was evaluated by observing clinical symptoms,detecting urine routine,serum creatinine(SCr),urea nitrogen(BUN),inflammatory factor-α(TNF-α),neutrophil gelatinase-related lipocalin(NGAL),kidney injury molecule-1(KIM-1).At the end of the experiment,renal tissue was taken for pathological examination.The results showed that compared with before modeling,gentamicin could lead to a significant increase in SCr and BUN,TNF-α,NGAL and KIM-1 also increased continuously,and abnormal urine routine.After AD-MSCs treatment,compared with the model group,the levels of SCr,BUN,TNF-αin the treatment group decreased,NGAL and KIM-1 returned to normal levels in the treatment group.The histological section revealed that the injury and necrosis of renal tubular epithelial cells in the treatment group were significantly lighter than those in the model group.These results indicated that intravenous injection of AD-MSCs had a certain therapeutic effect on canine AKI.
