能源甜菜转录因子BvMYB44基因响应镉胁迫的表达特性及生物信息学分析 被引量:2

摘要 为了解析BvMYB44在能源甜菜重金属胁迫中的调控作用,本研究以‘780016B/12优’为材料,通过qRT-PCR研究BvMYB44响应镉胁迫的表达模式,并利用生物信息学分析其功能。研究表明,BvMYB44基因cDNA全长942 bp,编码313个氨基酸,蛋白分子量为33935.01 Da,理论等电点为7.57。BvMYB44定位于细胞核中,具有39个磷酸化位点,为仅有1个外显子的不稳定碱性亲水蛋白。BvMYB44为R2R3-MYB亚家族成员,与藜麦CqMYB44-like和菠菜SoMYB44-like的亲缘关系最近。在该基因启动子区预测到多个与非生物胁迫相关的顺式作用元件,推测其可能参与多种非生物胁迫响应。qRT-PCR表明,在0.5 mmol/L镉胁迫处理下,BvMYB44在能源甜菜叶和根中均有不同程度的显著上调表达。因此,可以推断BvMYB44与镉胁迫存在着一定的应答关系。本研究将为抗重金属能源甜菜的种质创新以及拓展植物修复提供候选基因和理论基础。 In order to analyze the regulatory role of BvMYB44 of energy beet under heavy metal stress, this study used‘780016 B/12 You’as material to study the expression pattern of BvMYB44 in response to cadmium stress by qRT-PCR, and analyzed its function by bioinformatics. The results show that the total length of cDNA of BvMYB44 gene is 942 bp, encoding 313 amino acids, the molecular weight of the protein is 33935.01 Da,and the theoretical isoelectric point is 7.57. BvMYB44 is an unstable alkaline hydrophilic protein with 39 phosphorylation sites located in the nucleus and only one exon. BvMYB44 is a member of the R2 R3-MYB subfamily and is most closely related to CqMYB44-like from Chenopodium quinoa and SoMYB44-like from Spinacia oleracea. Several cis-acting elements related to abiotic stress are predicted in the promoter region of this gene, which is speculated to be involved in a variety of abiotic stress responses. The qRT-PCR shows that BvMYB44 has different levels of significantly up-regulated expression in energy beet leaves and roots under0.5 mmol/L cadmium stress. Therefore, it can be inferred that BvMYB44 has a certain response relationship with cadmium stress. This study will provide candidate genes and a theoretical basis for germplasm innovation of heavy metal-resistant energy beet and expansion of plant remediation.
