摘要
目的:筛选与儿童肥胖相关的血液中差异代谢物,探讨肥胖儿童代谢紊乱的发生机制。方法:采用气相色谱-质谱联用(GC/MS)和模式识别技术分析肥胖儿童血清中代谢物的变化,GC/MS检测25例肥胖儿童(肥胖组)和25例健康儿童(正常组)血清中代谢物,采用偏最小二乘判别分析(partial least squares discrimination analysis,PLS-DA)识别两组样本的血清代谢谱及代谢物。结果:肥胖组和正常组的代谢谱分离良好,肥胖儿童和正常儿童之间的血清代谢成分存在33个差异代谢物,绘制33个差异代谢物ROC曲线,计算曲线下面积,肌酐、马来酸、莽草酸等16个代谢物面积大于0.5,可能是诊断肥胖儿童潜在的生物标志物。结论:肥胖儿童患者血清代谢物的变化机制和甘氨酸,丝氨酸和苏氨酸的代谢、丙氨酸、天冬氨酸和谷氨酸代谢、乙醛酸和二羧酸酯代谢密切相关。
OBJECTIVE To screen for differential metabolites in blood related to childhood obesity and elucidate the mechanism of metabolic disorders in obese children.METHODS The changes in serum metabolites in obese children were examined by gas chromatography-mass spectrometry(GC/MS)and pattern recognition technology.GC/MS was utilized for detecting the serum metabolites of 25 obese children(obesity group)and 25 healthy controls(normal group).Partial least squares discrimination analysis(PLS-DA)pattern recognition was employed for identifying serum metabolic profiles and metabolites of two groups.RESULTS The metabolic profiles of obesity and normal groups were well separated.There were 33 different metabolites in serum metabolic components between obese and normal children.Receiver operating characteristic(ROC)curve of 33 different metabolites was plotted and area under the curve calculated.The area of 16 metabolites such as creatinine,maleic acid and shikimic acid was greater than0.5.It may be a potential biomarker for diagnosing pediatric obesity.CONCLUSION The mechanism of changes in serum metabolites in obese children is closely correlated with the metabolism of glycine,serine and threonine,the metabolism of alanine,aspartic acid and glutamate and the metabolism of glyoxylic acid and dicarboxylic acid esters.
