摘要
试验建立天冬糖苷散的质量控制方法,采用薄层色谱法定性鉴别天冬糖苷散中三七总皂苷,并采用高效液相色谱法测定天冬糖苷散中三七总皂苷含量,采用硫酸蒽酮法测定天冬糖苷散中总多糖的含量;进行碘-碘化钾反应、茚三酮反应、Molish反应、紫外光谱分析定性鉴别天冬糖苷散中的多糖。结果显示,天冬糖苷散与三七对照药材在薄层色谱图中相同位置有相同斑点,且斑点清晰无干扰,可有效鉴别三七总皂苷;高效液相色谱法可检测天冬糖苷散中三七总皂苷的含量,稳定可靠;硫酸蒽酮法测得天冬糖苷散中总多糖含量为59.90%;多糖的定性鉴别结果显示,碘-碘化钾和茚三酮反应呈阴性,Molish反应呈阳性,紫外光谱分析未见核酸和蛋白质。研究表明,试验建立的方法简便易行,稳定性和重复性较高,可有效地控制天冬糖苷散质量。
In order to establish the quality control method of Tian-Dong-Tang-Gan powder(TDTGP), TLC was used to qualitatively identify the total saponins of Panax notoginseng in TDTGP, the content of Panax notoginseng saponins in TDTGP was determined by high performance liquid chromatography, and the content of total polysaccharides in TDTGP was determined by the anthrone sulfate method. Iodine-potassium iodide reaction, ninhydrin reaction, Molish reaction,and ultraviolet spectroscopy are carried out to qualitatively identify polysaccharides in TDTGP. The results showed that TDTGP and Panax notoginseng control materials had the same spots in the same position in the TLC, and the spots were clear and non-interference, which could effectively identify the total saponins of Panax notoginseng. High performance liquid chromatography can effectively determine the content of total saponins of Panax notoginseng in TDTGP, which was stable and reliable. The content of total polysaccharides in TDTGP was 59.90% determined by anthrone sulfuric acid method. The results of physical and chemical properties showed that the reaction of iodine-potassium iodide and ninhydrin was negative, the Molish reaction was positive, and ultraviolet spectrum analysis showed no nucleic acid and protein. The experiment indicates that the method established in the experiment is simple and easy to implement, with high stability and reproducibility, and can effectively control the quality of TDTGP.
