黄芪多糖对大鼠慢性难愈合创面的作用及其对PTEN、AKT和VEGF蛋白表达的影响 被引量:18

Effects of Astragalus Polysaccharide on Rats’ Chronic Refractory Wounds and Protein Expressions of PTEN,Akt and VEGF

摘要 目的:观察黄芪多糖对慢性难愈合创面的促进愈合作用及其对同源性磷酸酶-张力蛋白(phosphatase and tension homology deleted on chromosome ten,PTEN)、丝-苏氨酸蛋白激酶(serine-threonine kinase,AKT)和血管内皮生长因子(vascular endothelial growth factor,VEGF)蛋白表达的影响。方法:建立慢性难愈合创面模型大鼠,随机分为模型组(0.5 g凡士林药膏)、黄芪多糖组(0.5 g黄芪多糖药膏)及京万红软膏组(0.6 g京万红软膏),同时设正常对照组(0.5 g凡士林药膏),各组每天给予相应外用药1次,连续给药3周。观察给药后7、14、21 d大鼠一般状态、体质量、伤口愈合率的变化,同时检测给药21 d后各组大鼠血清TNF-α、IL-6、VEGF含量变化。HE染色观察创面组织病理变化。Western blot检测创面组织PTEN、AKT、p-AKT、VEGF蛋白表达水平。结果:与正常对照组相比,模型组大鼠毛发无光泽,萎靡不振,摄食量减少,体质量显著降低(P <0.01),且血清TNF-α、IL-6水平显著升高(P <0.01)。与模型组相比,黄芪多糖组与京万红软膏组大鼠一般状态开始改善,体质量、创面愈合率显著增加(P <0.01),血清TNF-α、IL-6水平显著降低(P <0.05),VEGF含量显著升高(P <0.05)。HE染色结果显示给予黄芪多糖可减少炎性细胞浸润,促进血管、皮肤新生。Western blot检测结果显示黄芪多糖可下调创面组织PTEN蛋白表达(P <0.01),上调VEGF和p-AKT/AKT蛋白表达(P <0.01)。结论:黄芪多糖对慢性难愈合创面具有促进愈合作用,可通过抑制炎症反应,下调PTEN,上调VEGF和p-AKT/AKT蛋白表达,发挥促进血管生成、修复创面的作用。 Objective:To observe the healing promoting effect of Astragalus Polysaccharide on rats’ chronic refractory wounds and effects on protein expressions of PTEN,Akt and VEGF.Methods:To establish chronic refractory wounds model,rats were randomly divided into model group( 0.5 g petrolatum ointment),Astragalus Polysaccharide group( 0.5 g Astragalus Polysaccharide Ointment),and a Jingwanhong Ointment group( 0.6 g Jing wanhong Ointment),while normal control group( 0.5 g petrolatum ointment) was set,each group was given the corresponding Ointment for external application once a day for 3 weeks.The status,body weight,and wound healing rate of rats after 7,14,and 21 d of administration were observed,while the contents of serum TNF-α,IL-6,and VEGF after 21 d of administration were detected by ELISA.HE staining was performed to observe the pathological changes of wound tissues.The protein expression levels of PTEN,Akt,p-Akt and VEGF in wound tissues were detected by Western blot.Results:Compared with the normal control group,the rats in the model group had dull hair,malaise,decreased food intake,significantly lower body weight( P <0.01),and significantly higher levels of serum TNF-α and IL-6( P < 0.01).Compared with the modelgroup,the condition of rats in the Astragalus Polysaccharide group and the Jingwanhong Ointment group began to improve,with significant increases in bodyweight and wound healing rate( P < 0.01),the levels of TNF-α and IL-6 were significantly decreased( P <0.05),and the content of VEGF was significantly increased( P < 0.05).HE staining results showed that administration of Astragalus Polysaccharide could reduce inflammatory cell infiltration and promote angiogenesis and skin neogenesis.Western blot assay showed that Astragalus Polysaccharide could down regulate the protein expressions of PTEN( P < 0.01) and up regulate the protein expressions of VEGF and p-AKT/AKT( P < 0.01).Conclusion:Astragalus Polysaccharide could effectively promote chronic refractory wounds heal,which could play a role in promoting angiogenesis and repairing wounds by inhibiting the inflammatory response,down regulating the protein expression of PTEN and up regulating the protein expressions of VEGF and p-AKT/AKT.
