基于网络药理学探讨钩藤治疗抽动秽语综合征的作用机制 被引量:4

摘要 目的:基于网络药理学方法分析钩藤在治疗抽动秽语综合征方面的药理机制,为该中药在临床上的应用和推广提供参考和依据。方法:通过中药系统药理学数据库和分析平台获取钩藤的主要活性化学成分及其相关作用靶点;通过GeneCards、OMIM、TTD、GKB数据库获取抽动秽语综合征相关靶点,利用String平台进行蛋白质-蛋白质相互作用(Protein-Protein Interaction,PPI)分析,构建PPI网络并挖掘网络中潜在的蛋白质功能模块。在Bioconductor平台基础上运用R语言进行“药物-成分-靶点”分析,对其参与的生物过程及通路进行富集并将结果进行可视化,而后采用Cytoscape 3.8.0软件构建“钩藤成分-抽动秽语综合征靶点-通路”网络。结果:钩藤治疗抽动秽语综合征的核心活性成分为槲皮素(quercetin)、育亨宾(yohimbine)、柯楠因(Corynantheine)、毛钩藤碱(Hirsutine)、异去氢钩藤碱(coryincine)等,核心靶点有毒蕈碱型胆碱受体M1(Cholinergic Receptor Muscarinic 1,CHRM1)、雄激素受体(Androgen Receptor,AR),毒蕈碱型胆碱受体M3(Cholinergic Receptor Muscarinic 3,CHRM3)、阿片样物质受体δ1(Opioid Receptor Delta 1,OPRD1)、阿片受体Mu1(Opioid Receptor Mu 1,OPRM1)、肿瘤坏死因子(Tumor Necrosis Factor,TNF)、β2肾上腺素受体(Adrenoceptor Beta 2,ADRB2)等。钩藤调治抽动秽语综合征的生物学通路主要作用于神经活性配体-受体相互作用通路、疟疾、耶尔森氏菌感染、恰加斯病、类风湿关节炎、白细胞介素(Interleukin,IL)-17信号通路、AGE-RAGE信号通路(AGE-RAGE signaling pathway)等通路,其功能主要为影响神经递质释放等。结论:本研究初步揭示了钩藤调治抽动秽语综合征的多成分、多靶点、多通路的作用机制,为钩藤的临床用药提供基础。 Objective:To analyze the pharmacological mechanism of uncaria on tourette syndrome based on the network pharmacology method,and provide reference and basis for the clinical application and promotion of traditional Chinese medicine.Methods:The main active chemical components of uncaria and their related targets were obtained from the TCMSP database;the targets related to tourette syndrome were obtained from GeneCards,OMIM,TTD,and GKB databases,and the String platform performed PPI analysis,constructed a PPI network and excavated potential protein functional modules in the network.By the bioconductor platform,the R language was used to analyze the“drug-component-target”,the biological processes and pathways were enriched and the results were visualized.Then,Cytoscape 3.8.0 software was used to construct the“uncaria component-tourette”syndrome target-pathway network.Results:The core active ingredients of uncaria for the treatment of tourette syndrome were quercetin,yohimbine,corynin,rhynchophylline,coryincine,etc.The core targets were CHRM1,AR,CHRM3,OPRD1,OPRM1,TNF,ADRB2,etc..The biological pathway of uncaria for treating tourette syndrome mainly acts on the neuroactive ligand-receptor interaction,malaria,yersinia infection,chagas disease,rheumatoid arthritis,IL-17 signaling pathway,and AGE-RAGE signaling pathway,whose functions are mainly to affect nerves transmitter release,etc..Conclusion:This study has initially revealed the multi-component,multi-target,and multi-path mechanism of uncaria on tourette syndrome,which provides a basis for the clinical medication of uncaria.
