抗菌肽Melectin对急性败血症患者铜绿假单胞菌临床分离株的体外抗菌活性分析 被引量:1

摘要 目的研究Melectin对急性败血症临床分离株铜绿假单胞菌(PA)的体外抗菌活性。方法随机收集安徽理工大学第一附属医院急性败血症临床PA分离株,其中铜绿假单胞菌的多重耐药菌(Extreme-drugresistant Pseudomonasaeruginosa,MDRPA)10株,非耐药铜绿假单胞菌(Non-Extreme-drugresistantPseudomonasaeruginosa,Non-MDRPA)25株。应用微量肉汤稀释法检测Melectin对PA的体外最低抑菌浓度(minimum inhibitoryconcentration,MIC);选取MDRPA和Non-MDRPA的代表株各1株(MIC为16μg/mL),检测4×MIC浓度Melectin(64μg/mL)的杀菌动力学,并分别加入4,8,16μg/mLMelectin后,定时测定其600 nm处吸光度并绘制生长曲线。结果Melectin在体外条件下对MDRPA和Non-MDRPA均表现出良好的体外抗菌活性:Melectin抗MDRPA的MIC为16μg/mL,Non-MDRPA的MIC为16μg/mL,两组MIC差异无统计学意义(P>0.05)。4×MIC的Melectin在60 min时可以杀灭49.82%的MDRPA和53.33%的Non-MDRPA。体外培养24 h,Melectin浓度在低于MIC时(浓度为0、4、8μg/mL),MDRPA的生长基本不受影响。而Non-MDRPA在0、4μg/mL浓度下不受Melectin抑制,在半数MIC既8μg/mL浓度下,虽然0~15 h时细菌生长受到抑制,但在15 h后细菌呈快速生长趋势。在MIC浓度(16μg/mL)Melectin进行作用时,MDRPA和Non-MDRPA均不再生长。结论Melectin体外条件下可以有效抑制PA急性败血症临床分离株的生长,有望发展成为一种新型抗菌药物。 Objective To investigate the in vitro antibacterial activity of Melectin against Pseudomonas aeruginosa(PA),a clinical isolate of acute sepsis.Methods Ten multi-drug resistant Pseudomonas aeruginosa(MDRPA)isolates were randomly collected from the First Affiliated Hospital of Anhui University of Technology,and 10 non-drug resistant Pseudomonas aeruginosa(Non-Extreme-drug resistant PA)isolates were collected.The test was carried out using a mi⁃cro-broth dilution method for the detection of Mead Johnson.The in vitro minimum inhibitory concentration(MIC)of Melectin against PA was determined by micro-broth dilution method;one representative strain of MDRPA and one rep⁃resentative strain of Non-MDRPA(MIC of 16μg/mL)were selected,and the bactericidal kinetics of 4×MIC concentra⁃tion of Melectin(64μg/mL)were determined.The kinetics of fungicidal activity of 4×MIC concentration of Melectin(64μg/mL)was measured,and the absorbance at 600 nm was measured at regular intervals after the addition of 4,8 and 16μg/mL of Melectin,and the growth curves were plotted.Results The micro-broth dilution method showed that for both the MDRPA and non MDRPA the MIC was the same(16μg/mL).Melectin at the concentration of 4×MIC killed 49.82%of MDRPA and 53.33%of Non-MDRPA 60 min after the addition of the drug..The growth of MDRPA was basically not affected by the Melectin at the MIC concentration;and the non-MDRPA was not inhibited by Melec⁃tin at the concentration under 4μg/mL,and was inhibited by Melectin at the concentration of 8μg/mL for 0~15 h,how⁃ever,the bacteria grew fast after 15 h.When the Melectin concentration was≥MIC(16μg/mL),both MDRPA and non MDRPA ceased to grow.Conclusion Melectin effectively inhibits the growth of PA under in vitro condition and is ex⁃pected to become a novel antibacterial drug.
