摘要
土地盐碱化是全球关注的环境问题之一。中国盐碱地分布广、面积大,筛选具有耐盐性的紫花苜蓿品种可有效利用盐碱地。该研究选取33个品种紫花苜蓿种子,分别在0(CK)、0.3%、0.6%、0.9%、1.2%和1.5%NaCl溶液处理下进行萌发试验,测定发芽势、发芽率和胚根长、幼苗长等指标,通过聚类、单因素和一元二次方程分析,对33个品种进行耐盐性分级并确定理论鉴别浓度。结果表明:随着盐胁迫增强,22份苜蓿种质材料相对发芽率、相对发芽势、相对根长和相对芽长较对照均显著降低(P<0.05),仅有部分苜蓿品种的种子表现出低盐浓度促进种子萌发和幼苗生长现象,在受到盐胁迫时,根较芽更敏感;苜蓿半致死盐浓度(S_(50))均值1.24%,为耐盐性鉴定的适宜浓度;由聚类分析可得,WL198HQ和WL903两个品种在萌发期具有较强的耐盐性,皇后、可汗等12个品种对盐胁迫较为敏感。该研究结果可为耐盐苜蓿新品种选育提供参考。
Land salinization is one of the environmental issues of global concern.Saline-alkali land is widely distributed in China,Screening alfalfa cultivars with salt tolerance can effectively utilize saline-alkali land.In the present study,the seeds of 33 alfalfa cultivars were germinated and cultured in CK,0.3%,0.6%,0.9%,1.2%and 1.5%NaCl solution for 10 days.The germination potential,germination rate,root length and shoot length were measured.Through cluster analysis,single factor analysis and quadratic equation analysis,the salt tolerance of 33 cultivars was graded and the theoretical identification concentration was determined.With the increase of salt concentration,the relative germination rate,relative germination potential,relative root length and relative sprout length of 22 alfalfa germplasm materials were significantly reduced compared with the control,but a few alfalfa varieties showed better performance with seed germination and seedling growth in low salt concentration.And alfalfa roots are more sensitive to salt than buds.The average half-lethal salt concentration(S_(50))of alfalfa is 1.24%,which is the appropriate concentration for salt tolerance identification,theoretically.Cluster analysis showed that WL198HQ and WL903 are two salt-tolerance varieties during the germination period,and 12 salt-sensitive varieties include Queen and Khan,etc.The research results can provide references for the selection and breeding of new salt-tolerant alfalfa varieties.
