摘要
对高效毛细管电泳法(HPCE)测定辛弗林含量的条件进行了优化,并采用优化后的HPCE法测定了沅江酸橙果实中辛弗林的含量,为其质量评价、控制和最佳采收期确定等提供参考。方法优化结果表明:辛弗林含量在33~530 μg/mL的浓度范围内呈良好线性关系,相关系数R^(2)为0.999 9,精密度、重复性、稳定性试验结果的相对标准差(RSD)均小于3.0%,平均加样回收率为93%,RSD为2.3%;该测定方法简单、高效、准确,适用于沅江酸橙中辛弗林的含量测定。酸橙样品辛弗林含量测定结果表明:采收期对酸橙果实中辛弗林含量有显著性影响(P<0.05),沅江的枳实以5月至6月中旬采收为宜,枳壳以7月中旬到8月中旬采摘为宜;第一批(即5月18日采摘)21株酸橙种质辛弗林含量为7.64~9.96 mg/g,不同种质间含量差异显著(P<0.05),其中编号为9、21、50、65、77的酸橙辛弗林含量最高,可为高生物碱特性的酸橙品种选育提供优异的种质资源。
In this study,the HPCE method was optimized and used for the determination of synephrine content in Yuanjiang Citrus aurantium fruit samples to provide reference for quality evaluation and control,and determination of the optimal harvest time.The results showed that the content of synephrine had a good linear relationship in the concentration range of 33-530 μg/mL,and the correlation coefficient R^(2) was 0.999 9.The RSD of precision,repeatability and stability each was less than 3.0 %.The average recovery rate was 93 %,and the RSD was 2.3 %.The method is simple,efficient and accurate,which is suitable for the determination of synephrine content in Yuanjiang Citrus aurantium fruit.Drawn from the determination results,the harvest time had a significant effect on the content of synephrine in Citrus aurantium fruit (P < 0.05).The optimal harvest time should be from May to mid June for the immature fruit of Yuanjiang Citrus aurantium,and from mid July to mid August for the mature fruit.The content of synephrine in 21 Citrus aurantium germplasms (harvested on May 18th) ranged from 7.64 to 9.96 mg/g,and the content of synephrine in code 9,21,50,65 and 77 Citrus aurantium germplasms was higher,which could provide excellent germplasm resources for breeding Citrus aurantium varieties with high alkaloid characteristics.
