当归芍药散保护大鼠子宫缺血再灌注损伤的影响机制探讨 被引量:2

Study on the Mechanism of the Effect of Danggui Shaoyao Powder in the Protection of Uterine Ischemia-reperfusion Injury in Rats

摘要 目的:观察当归芍药散对大鼠缺血再灌注损伤模型子宫的影响并分析其可能机制。方法:将30只雌性SD大鼠按随机原则均分为假手术组、模型组和实验组,每组10只,除假手术组外均用无创动脉夹夹闭大鼠子宫圆动脉30 min后再灌注60 min以构建大鼠子宫缺血再灌注模型。术后2 h即灌胃给予实验组当归芍药散[1.35 mg/(kg·d)],另2组均灌胃给予等体积0.9%氯化钠溶液作为对照。于治疗7 d后取大鼠子宫组织检测高能磷酸化合物及子宫缺血再灌注损伤相关酶学指标,并结合子宫活动力评价其对缺血再灌注损伤模型子宫能量代谢及生理功能的影响。结果:实验组子宫收缩频率、收缩力及子宫活动力均较模型组明显降低,且高能磷酸化合物总腺嘌呤核苷酸(total adenine nucleotides,TAN)、二磷酸腺苷(adenosine diphosphate,ADP)、三磷酸腺苷(adenosine triphosphate,ATP)、磷酸腺苷(adenosine monophosphate,AMP)水平及超氧化物歧化酶(superoxide dismutase,SOD)均明显高于模型组,而乳酸脱氢酶(lactate dehydrogenase,LDH)和丙二醛(malondialdehyde,MDA)水平明显低于模型组,差异有统计学意义(P<0.05)。结论:当归芍药散可降低子宫活动力,对子宫缺血再灌注损伤具有保护作用,这一作用可能是通过影响高能磷酸化合物代谢、增强缺血再灌注子宫组织能量供应并抑制氧化应激反应而实现的。 Objective To observe the effect of Danggui Shaoyao powder on the uterus of rats with ischemia-reperfusion injury and analyze its possible mechanism.Methods Thirty female SD rats were randomly divided into sham operation group,model group and experimental group,with 10 rats in each group.Except for the sham operation group,the uterine circular artery was clamped with non-invasive arterial clips for 30 min and then reperfusion for 60 minutes to construct the uterine ischemia-reperfusion model of rats.The experimental group was given Danggui Shaoyao Powder[1.35 mg/(kg·d)]by gavage 2 hours after operation,and the other two groups were given equal volume 9%sodium chloride solution as the control.After 7 days of treatment,the uterine tissue of rats was taken to detect the high-energy phosphate compounds and the enzyme indexes related to uterine ischemia-reperfusion injury,and its effects on uterine energy metabolism and physiological function of ischemia-reperfusion injury model were evaluated in combination with uterine activity.Results The uterine contraction frequency,contractile force and uterine activity in the experimental group were significantly lower than those in the model group,and the high-energy phosphate compound total adenine nucleotides(TAN),adenosine diphosphate(ADP),adenosine triphosphate(ATP),adenosine monophosphate(AMP)levels and superoxide dismutase(SOD)were significantly higher than those in the model group,while the levels of lactate dehydrogenase(LDH)and C the level of malondialdehyde(MDA)were significantly lower than those in the model group,and the difference between the groups was statistically significant(P<0.05).Conclusion Danggui Shaoyao powder can reduce uterine activity and protect against uterine ischemia-reperfusion injury.This effect may be achieved by affecting the metabolism of high-energy phosphate compounds,enhancing the energy supply of ischemia-reperfused uterine tissue and inhibiting oxidative stress.
