Lipid metabolism characteristics of cancer stem cells

摘要 肿瘤干细胞(cancer stem cells, CSCs)是肿瘤中能够自我更新和致瘤的一小群异质性细胞。它们是肿瘤发生、转移、复发和耐药的潜在动力。目前研究发现,CSCs的干性维持很大程度上依赖于它特有的脂质代谢改变,它们可以通过各种手段来维持干性和自身环境的稳态,如:提高胞内脂肪酸(fatty acids, FA)的含量来供能、促进FA的β氧化来增强能量利用率、上调甲羟戊酸途径来合成胆固醇等。此外,脂滴(lipid droplets, LDs)可以在CSCs糖酵解受阻时提供代替能源,保护FA免受过氧化的损害;自噬与脂质代谢的相互作用使CSCs能够迅速适应化疗后产生的恶劣微环境。本文对近年来CSCs脂质代谢相关的研究进行了综述,简要介绍了LDs、FA和胆固醇代谢以及自噬对于CSCs干性维持的必要性。 Cancer stem cells(CSCs)are small group of heterogeneous cells in tumors that are capable of self-renewal and tumorigenesis.They are the underlying drivers of tumor genesis, metastasis, recurrence and drug resistance.At present, the study found CSCs stemness maintenance largely depends on its characteristic of lipid metabolic changes, they can through various means to maintain stemness and its microenvironment, such as: improve the intracellular content of fatty acid(FA)to supply energy, promote the FA β-oxidation to enhance energy efficiency, increase mevalonate pathway to improve synthesis of cholesterol, etc.In addition, lipid droplets(LDs)can provide alternative energy to protect FA from peroxidation when the glycolysis of CSCs is blocked.The interaction between autophagy and lipid metabolism enables CSCs to quickly adapt to the harsh microenvironment generated by chemotherapy.In this review, we reviewed recent studies on lipid metabolism of CSCs, and briefly introduced the necessity of LDs, FA, cholesterol metabolism and autophagy for stemness maintenance of CSCs.
