摘要
目的:对比研究不同剂量的红景天破壁饮片对小鼠的急性肝肾毒性。方法:将40只清洁级昆明小鼠随机分成空白组对照组、红景天破壁饮片低、中、高剂量组,每组10只,雌雄各半,禁食不禁水12h后,分别给予红景天破壁饮片低、中、高剂量(29.578mg/g、34.797mg/g、40.938mg/g),空白组给于相同体积的蒸馏水,每天灌胃3次。连续给药14天后,观察小鼠一般体征及各脏器指数;检测小鼠血清中谷草转氨酶(AST)、碱性磷酸酶(ALP)、白蛋白(ALB)、尿素氮(BUN)、血糖(Glu)、总胆固醇(TC)及甘油三酯(TG)的含量;HE染色观察小鼠肝肾组织病理变化。结果:与空白组比较,红景天破壁饮片各剂量组小鼠血清中谷草转氨酶(AST)、碱性磷酸酶(ALP)、白蛋白(ALB)、尿素氮(BUN)、血糖(Glu)、总胆固醇(TC)、甘油三酯(TG)含量及小鼠脏器指数差异均不显著(P>0.05);与空白组比较,各给药剂量组肝肾组织HE染色差异均不显著(P>0.05)。结论:红景天破壁饮片不会引起小鼠机体肝肾等组织的毒副反应,安全性较高。
Objective:Comparative study on the acute Hepatic and kidney toxicity of different doses of Rhodiola decoction pieces on mice.Methods:Forty clean Kunming mice were randomly divided into a blank group,a control group,and a low medium,and high-dose Rhodioladecoction pieces.Each group had 10 mice,half male and half.After 12 hours of fasting and water,they were given to Yujing.The low,medium and high dose groups of Rhodiola broken decoction pieces(29.578 mg/g,34.797 mg/g,40.938 mg/g)and the blank group were given the same volume of distilled water.Gavage the stomach 3 times a day.After 14 days of continuous administration,observe the general physical signs and various organ indexes of the mice;detect the aspartate aminotransferase(AST),alkaline phosphatase(ALP),albumin(ALB),urea nitrogen(BUN),blood glucose(Glu),total cholesterol(TC)and triglyceride(TG)content;HE staining to observe the pathological changes of mouse liver and kidney tissue.Results:Compared with the blank group,aspartate aminotransferase(AST),alkaline phosphatase(ALP),albumin(ALB),urea nitrogen(BUN),blood glucose(Glu)and total serum levels of mice in each dose group of Rhodiolabroken decoction pieces There was no significant difference in cholesterol(TC),triglyceride(TG)content,and organ index of mice(P>0.05);compared with the blank group,the difference in HE staining of liver and kidney tissue in each dose group was not significant(P>0.05).Conclusion:Rhodiolabroken wall drink will not cause toxic and side effects in mice’s liver and kidney and other tissues,and it is safer.
