SD大鼠癫痫发作后海马区Wnt3a、p-β-catenin和 β-catenin的表达变化 被引量:1

The Changes of Hippocampal CA3 Region Wnt3a,p-β-catenin andβ-catenin Proteins Expression in SD Epileptic Rats

摘要 目的探讨氯化锂-匹罗卡品致SD大鼠癫痫发作后,在不同时期海马区Wnt3a、p-β-catenin及β-catenin表达的变化。方法将32只SD大鼠随机分为正常对照组(Con组)、癫痫模型组(SE 1 d、SE 7 d和SE 14 d)共4组,每组8只。癫痫模型组大鼠应用腹腔注射氯化锂-匹罗卡品成功制备SD大鼠癫痫模型,通过水迷宫实验观察各组大鼠的学习记忆情况。应用Western blot测定癫痫发作后各组海马区GFAP、Wnt3a、p-β-catenin及β-catenin的表达情况。结果水迷宫实验结果显示,与Con组比较,SE 7 d组、SE 14 d组大鼠第2~5天逃避潜伏期延长、60 s内定位巡航路程延长、穿越平台的次数减少(P均<0.05);SE 7 d组首次穿越平台的时间长于Con组(P<0.05)。蛋白电泳结果显示,SE 7 d组和SE 14 d组癫痫大鼠海马区GFAP、Wnt3a、β-catenin蛋白水平均较Con组升高(P均<0.05),SE 7 d组和SE 14 d组大鼠p-β-catenin蛋白表达量均低于Con组(P均<0.05)。结论SD大鼠癫痫发作后,学习记忆能力下降。海马区GFAP、Wnt3a、β-catenin蛋白表达量升高,且在癫痫的早期,随时间的延长有上调的趋势。 Objective To investigate the changes of hippocampal Wnt3a,p-β-catenin andβ-catenin proteins expression at different times after epileptic seizure in lithium-pilocarpine induced epileptic rats.Methods 32 adult male SD rats were divided into control(Con)group and SE 1 d,SE 7 d,SE 14 d groups,which are eight rats in each group.The SD rats of epileptic model groups were successfully prepared by intraperitoneal injection of lithium-pilocarpine.The learning and memory abilities of epilepstic model rats in each group were observed and analyzed by morris water maze test,and the levels of hippocampal GFAP,Wnt3a,p-β-catenin andβ-catenin proteins expression in each group were detected by Western blot protein analysis.Results The results of the morris water maze test showed that,compared with the Con group,the rats in the SE 7 d and SE 14 d groups had longer latency to evade on days 2-5,longer localization cruising distance within 60 s,and fewer times to cross the platform(P all<0.05),the time to cross the platform for the first time was longer in the SE 7 d group than in the Con group(P<0.05).The protein electrophoresis results showed that the levels of GFAP,Wnt3a andβ-catenin in the hippocampal were higher in the SE 7 d and SE 14 d groups than in the Con group(P all<0.05),and the expression of p-β-catenin in the SE 7 d and SE 14 d groups was lower than that in the Con group(P all<0.05).Conclusion The learning and memory abilities of SD rats significantly decreased after epileptic seizure.The levels of hippocampal GFAP,Wnt3a andβ-catenin proteins expression increased,and which showed a tendency of up-regulation with the extension of time in the early stage of epilepsy.
