miRNA-506在三阴性乳腺癌中的表达及对其增殖和侵袭作用的研究 被引量:1

The expression of miRNA-506 in triple-negative breast cancer and its effects on proliferation and invasion of the cancer cells

摘要 目的探讨miRNA-506对三阴性乳腺癌癌细胞增殖及侵袭能力的影响。方法qRT-PCR检测三阴性乳腺癌患者癌组织、癌旁正常组织、乳腺良性病变组、三阴性乳腺癌细胞株MDA-MB-231以及正常乳腺上皮细胞系HBL-100中miRNA-506表达情况;转染miRNA-506mimics后CCK-8检测细胞增殖情况;Transwell小室法检测MDA-MB-231细胞侵袭情况。结果乳腺癌组织中的miRNA-506表达水平低于癌旁正常组织以及乳腺良性病变组织(P<0.01);人三阴性乳腺癌细胞株MDA-MB-231细胞内的miRNA-506低于人正常乳腺上皮细胞系HBL-100细胞(P<0.01);转染miRNA-506mimics可显著抑制MDA-MB-231细胞增殖和侵袭(P<0.01)。结论miRNA-506在三阴性乳腺癌低表达以及其可抑制三阴性乳腺癌的增殖、侵袭生物学行为。 Objective To investigate the effects of miRNA-506 on the proliferation and invasion of triple-negative breast cancer cells.Methods qRT-PCR was used to detect the expressions of miRNA-506 in cancer tissues,normal adjacent tissues,tissues with benign breast lesions,as well as in triple-negative breast cancer cell line MDA-MB-231 and normal breast epithelial cell line HCL-100 of patients with triple-negative breast cancer.Cell proliferation was detected by CCK-8 after transfection of miRNA-506 mimics.The invasion of MDA-MB-231 cells was detected by Transwell cell assay.Results The expression of miRNA-506 in breast cancer tissues was lower than that in adjacent normal tissues and benign breast lesions(P<0.01).The expression of miRNA-506 in triple-negative breast cancer cell line MDA-MB-231 was lower than that in normal breast epithelial cell line HBL-100(P<0.01).Transfection of miRNA-506 mimics significantly inhibited the proliferation and invasion of MDA-MB-231 cells(P<0.01).Conclusion miRNA-506 has low expression in triple negative breast cancer and can inhibit the biological behavior of proliferation and invasion of triple-negative breast cancer.
